Serum Biomarker Profiling in Mice

Fasting Blood Analysis in Mice

Before the end of the study, at weeks six and twelve, the mice were fasted for six hours in the morning. Under isoflurane anesthesia, blood was collected via cardiac punction, and euthanasia was performed by cervical dislocation. Blood samples were incubated for 30 min at room temperature and centrifuged for 15 min at 1500× g, and the obtained serum was immediately frozen and stored at −80 °C. Serum cholesterol and triglyceride levels as well as alanine aminotransferase (ALT) activity were measured by fluorometric assays (Abcam, Cambridge, UK). Serum insulin (Mercodia Ltd., Uppsala, Sweden), serum amyloid A (SAA), leptin, resistin, adipsin, and adiponectin (R&D Systems Europe Ltd., Abingdon, UK) were measured by enzyme-linked immunoassays (ELISAs). The detection limits were 33 pmol/L for insulin, 62.5 ng/L for SAA, 7.8 ng/L for leptin and resistin, 375 ng/L for adipsin, and 15.6 ng/L for adiponectin.

Plasma Protein Quantification Protocol

Plasma samples were diluted in PBS (phosphate-buffered saline) 100 times. The same amount of diluted plasma sample was loaded for western blotting analysis. Antibodies of Adipsin (R&D Systems, Minneapolis, MN, USA; catalogue AF5430) and Adiponectin (Thermo Fisher Scientific, Waltham, MA, USA, catalogue PA1-054) were used in this study. The band intensity was quantified using ImageJ (v. 2.1.0; National Institutes of Health).

Metabolic Profiling in Diet-Induced Obesity

Every 2 weeks, the 12 h fasting blood glucose level was measured from tail vein blood with a glucose analyzer (GlucoDr SuperSensor, Allmedicus, Anyang, Korea). For the IPGTT analysis performed in the 15th week after the start of the diet experiments, mice fasted for 12 h were injected intraperitoneally with glucose (0.5 g/kg body weight), and the blood glucose level was determined in the tail vein blood at 0, 30, 60, and 120 min later. Plasma biomarkers were measured using fluorescent magnetic bead-based assays (Bio-Rad, Millipore, Hercules, CA, USA): insulin, glucagon, leptin, resistin, plasminogen activator inhibitor-1 (PAI-1), glucagon-like peptide-1 (GLP-1) and gastric inhibitory polypeptide (GIP) (Bio-Plex Pro™ mouse diabetes assays), IL-6, TNF-α, monocyte chemoattractant protein-1 (MCP-1) and macrophage inflammatory protein 1 beta (MIP-1β) (Bio-Plex Pro™ mouse cytokine assays), and adiponectin (Bio-Plex Pro™ mouse diabetes adiponectin assay). The plasma adipsin (R&D Systems, Minneapolis, MN, USA), soluble CD antigen 163 (sCD163; Mybiosource, San Diego, CA, USA), glutamic oxaloacetic transaminase (GOT) (Asan, Seoul, Korea), and glutamic pyruvic transaminase (GPT) (Asan, Seoul, Korea) levels were analyzed using a commercially available kit.

Complement C3a/C5a Signaling Assays

Recombinant C3a and C5a proteins (R&D) were used at a concentration of 100 nM. Adipsin (R&D) and C3 (Calbiochem) proteins were used at 1 µg/ml. Anti-Adipsin antibody (Santa Cruz) was used for Western blot analysis. Antibodies to C3aR1 (Santa Cruz), C5aR1 (Biolegend) and C5L2 (Hycult) were used for flow cytometry. RNA isolation and reverse transcription were performed with Trizol (Life Technologies), RNeasy (Qiagen) and cDNA synthesis (Agilent) kits. qPCR reactions were done with SYBR green on an ABI 7900HT (Life Technologies).
