Approximately 106 PSMA+ CWR22Rv1 cells in 50/50 complete medium and matrigel matrix were implanted in the right shoulder of athymic NCr-nu/nu male mice (approximately 8 weeks old) from Charles River (Hollister, CA). Four weeks after implantation, mice with tumors reaching 150 to 300mm3 were anesthetized by isoflurane inhalation and injected with [18F]5, 150–200 μCi in 250 μL of saline, in a tail vein. The resulting animals were imaged with 10 min acquisition by a microPET/CT imaging system (Inveon, Siemens, Germany) at 2 h postinjection. PET imaging data were acquired in list mode and reconstructed with the iterative OSEM 2-D reconstruction algorithm provided by the manufacturer.
