Cardiac Troponin I Detection Assay

Goat anti-mouse immunoglobulin G was obtained from Jackson ImmunoResearch Laboratories, Inc. (West Grove, PA, USA). Cardiac troponin I monoclonal antibody (4T21cc–19C7cc) and cardiac troponin I monoclonal antibody (4T21cc–560) were purchased from HyTest Ltd. (Turku, Finland). Cardiac troponin I-C complex was obtained from MyBioSource, Inc. (San Diego, CA, USA). Albumin, bilirubin, cholesterol, gold (III) chloride trihydrate (HAuCl4•3H2O), human serum, phosphate buffered saline (PBS), silver nitrate and Tween 20 were purchased from Sigma-Aldrich (St. Louis, MO, USA). Bovine serum Albumin (BSA) was purchased from Capricorn Scientific GmbH (Ebsdorfergrund, Germany). Hydroquinone was purchased from Tokyo Chemical Industry Co., Ltd. (Tokyo, Japan). Citric acid monohydrate and sodium citrate dihydrate were obtained from Bio Basic Inc. (Ontario, Canada). All analytical-grade chemicals and reagents, and mill-Q (18 MΩ-cm) water were used in the experiments.

Synthesis and Characterization of Compound 10-5

Added to sulfuric acid (1.84 mL) were a mixture of 2-(4-diethylamino-2-hydroxybenzoyl)-4-methyl-benzoic acid and 2-(4-diethylamino-2-hydroxybenzoyl)-5-methyl-benzoic acid (compound (9-5), 0.562 mmol, synthesized by a method described in document: WO2014/002292); and hydroquinone (0.280 mmol, purchased from Tokyo Chemical Industry Co., Ltd.), followed by stirring them at 110° C. for 4.5 hours. The reaction solution was then neutralized with an aqueous sodium hydroxide solution, and an insoluble(s) was later collected by filtration. The insoluble(s) was dissolved in chloroform, and then purified by silica gel chromatography to obtain a compound (10-5) as a faint blue solid. A yield was 7%. The reaction formula is shown below.

UV/Vis: λmax=659 nm (dicationic form, 10 vol % CF3COOH/CHCl3).

HRMS (ESI) m/z calcd. for C46H37N2O6 ([M+H]+): 693.2959, found: 693.2978.

Synthesis and Characterization of Anion-Exchange Membranes

NaOH and NH4PF6 were purchased from Fujifilm Wako Chemicals Co. HCl and Na2WO4·2H2O were obtained from Kanto Chemical Co. 2,2,6,6-Tetramethyl-4-piperidyl methacrylate, 3-[[2-(methacryloyloxy)ethyl]dimethylammonio]propane-1-sulfonate, thioglycolic acid, 4,4′-azobis(4-cyanovaleric acid), hydrogen peroxide, ethylviologen dibromide, 4,4′-bipyridyl, indomethane, 2-bromoethanol, methacryloyl chloride, triethylamine, hydroquinone and tetrabuthylammonium chloride were purchased from Tokyo Chemical Industry Co. Other chemicals were obtained from the above companies. All compounds were used as received. The anion-exchange membrane was obtained from Takahata precision Co. The membrane originally had methanesulfonate anions. Chloride polymers were obtained after immersing them in 1 M NaCl aqueous solution at 80 °C for 2 days. For control experiments, Nafion perfluorinated membrane (product number: 676470, thickness 0.002 inch) was purchased from Sigma-Aldrich Co., and SELEMION (anion-exchange membrane, product name: AMVN, chloride anions) was obtained from AGC Engineering Co. Nafion was immersed in 0.1 M NaOH aqueous solution overnight for neutralization.

Cortisol Immunoassay with Gold Nanoparticles

A mouse anti-cortisol monoclonal antibody, cortisol-BSA conjugate was purchased from Cusabio Biotech Co., Ltd, (China). Goat anti-mouse immunoglobulin G was obtained from Jackson ImmunoResearch Laboratories, Inc., Ethanol, disodium hydrogen orthophosphate, and sodium dihydrogen phosphate were purchased from BDH Prolabo (Lutterworth, UK). Hydrocortisone, 20 nm gold nanoparticles, citric acid, disodium citrate tribasic dehydrate, bovine serum albumin (BSA), and silver nitrate were purchased from Sigma-Aldrich (Missouri, USA). Hydroquinone was provided by Tokyo Chemical Industry Co., Ltd. Analytical-grade reagents and 18 MΩ-cm water were used throughout this experiment. The silver enhancer solutions were prepared by a 1:1 (V:V) mixing solution of 100 mg silver acetate in 50 mL milli Q water and 250 mg Hydroquinone in a 50 mL of citrate buffer pH 3.8.
