ChIP-qPCR Analysis of ZEB1 in PDAC

ChIP experiments were performed using 1 × 107 pancreatic cancer cells with the ChIP kit (Cell Signaling Technology) according to the manufacturer’s protocol. Briefly, PDAC cells were cross-linked with 1% formaldehyde. Then the cells were lysed and chromatin was harvested and fragmented using enzymatic digestion. The chromatin was immunoprecipitated using ChIP-grade antibody against ZEB1 (Sigma, United States). After immunoprecipitation, the protein-DNA cross-links were reversed and the DNA was purified, followed by qRT-PCR.
