Polymersomes Synthesis and Characterization

Chemicals were used as received unless otherwise indicated. 2-(Methacryloyloxy)ethyl phosphorylcholine (MPC > 99%) was donated by Biocompatibles. 2-(Diisopropylamino)ethyl methacrylate (DPA) was purchased from Scientific Polymer Products. Copper(I) bromide (CuBr; 99.999%), 2,2′-bipyridine (bpy), methanol (anhydrous, 99.8%), and isopropanol were purchased from Sigma-Aldrich. The silica used for the removal of the atom transfer radical polymerization (ATRP) copper catalyst was column chromatography grade silica gel 60 (0.063 to 0.200 mm) purchased from E. Merck. 2-(N-morpholino)ethyl 2-bromo-2-methylpropanoate (ME-Br) initiator was synthesized according to a previously reported procedure (50 ). Poly(ethylene glycol) methyl ether methacrylate [P(OEG10MA)] was purchased from Sigma-Aldrich. PEO-PBO copolymer was purchased from Advanced Polymer Materials Inc. The polymersomes were labeled using rhodamine B octadecyl ester perchlorate purchased from Sigma-Aldrich. PBS was made from Oxoid tablets (one tablet per 100 ml of water). Bovine liver catalase, glucose oxidase, and glucose have been purchased from Sigma-Aldrich. The gel filtration column for the purification of the polymersomes was made with Sepharose 4B purchased from Sigma-Aldrich.

Synthesis and Characterization of PMMA-SO3H Polymers

Poly(methyl methacrylate-co-3-sulfopropyl methacrylate) polymers (PMMA-SO3H 1 or 2 %) were synthetized through free radical polymerization as described previously. [11, 12, 43] R18/F5-TPB was synthesized from rhodamine B octadecyl ester perchlorate (Sigma-Aldrich, >98.0%) and lithium tetrakis(pentafluorophenyl)borate ethyl etherate (Alfa Aesar, 97%) through ion exchange followed by purification through column chromatography according to the method previously described. [38, 45] Milli-Q water (Millipore), NaCl (≥98%, Sigma-Aldrich), acetonitrile (≥99.9%, Sigma-Aldrich) and Tween 80 (T80, Polysorbate 80, Sigma-Aldrich) were used for preparation of NPs.

Lipid-Based Nanoparticle Preparation
