Fluorescence-based Enzymatic Assay

Singlet Oxygen Detection in Cu-Cy Nanoparticles

A singlet oxygen sensor green reagent (SOSG) (Ex = 505 nm, Em = 525 nm) was used to detect the 1O2 in the aqueous solution. SOSG (100 μg, Meilunbio, China) was dissolved in methanol (33 μL) to make a 5 mM stock solution. The SOSG stock solution was diluted with PBS to a working solution of 50 μM. 100 μL of Cu-Cy nanoparticles (10, 20, 40, and 80 μg/mL) were loaded into four wells of a 96-well plate, and 10 μL of SOSG stock solution was added to each well. After mixing, MW (20 W, 3 min) was irradiated, and the fluorescence of SOSG was measured with a microplate reader immediately after each microwave treatment.

DHE and cell culture products were purchased from Keygentec (Nanjing, China). All the reagents, except to that were not pointed out specially, were analytical grade, and ultra-pure water was used in the preparation of all samples.
