CD44 Expression Analysis by Flow Cytometry

CD44 Expression in TS/A-pc and HeLa Cells

To measure expression of the HA-binding receptor CD44 in TS/A-pc and HeLa cells, TS/A-pc and HeLa cells were seeded into a 6well plate at a density of 5 × 10 5 cells per well and incubated for 24 h at 37 °C in a humidified atmosphere of 5% CO2. Cells were trypsinated, counted and washed before incubation for 1 h in 300 µL of PBS containing 10 µL of fluorescein isothiocyanate (FITC)conjugated mouse anti-human CD44 antibody (#555478 from BD Pharmingen) at 4 °C. FITC mouse IgG2b κ antibody was used as isotype control (# 555742 from BD Pharmingen). After three washing with PBS, the cells were observed using fluorescence-activated cell sorting (FACS) Accuri 6 (Becton, Dickinson and Company, Pont de Claix, France). Cells were gated to exclude cell debris and 10,000 events were recorded in the gate for fluorescence intensity measurement.
