Quantitative Real-Time PCR Procedure

Cardiac Gene Expression Quantification

Total RNA was isolated from the heart tissues using the RNAiso Plus according to the manufacturer's instruction (Takara, Tokyo , Japan). Five hundred nanograms of total RNA was reverse transcribed into complementary DNA in a 10 μl reaction mixture using the PrimeScriptTM RT reagent kit (Takara, Tokyo, Japan). The Q-PCR was performed using the SYBR Green PCR Master Mix (Takara, Tokyo, Japan) and the ABI Q6 Flex Real-Time PCR machine (Applied Biosystems, Foster City, CA, USA). The relative gene expression levels were analyzed using the 2(-ΔΔCt) method and normalized against GAPDH expression. The primers used are listed in Table S2.
