Colorectal Cancer Cell Line Maintenance

The human colorectal cancer cell lines used in this study were purchased from ATCC (Manassas, VA). The CCD-18Co normal colon fibroblast cell line was purchased from the Korea Cell Line Bank (KCLB) (Seoul, Korea). The HCT15, SW620, SW480, SW48, LoVo, and DLD-1 cells were maintained in RPMI 1640 medium supplemented with 10% fetal bovine serum, 100 U/mL penicillin, and 100 μg/mL streptomycin. The HT29 and HCT116 cells were maintained in McCoy’s 5A medium supplemented with 10% fetal bovine serum, 100 U/mL penicillin, and 100 μg/mL streptomycin. The CCD-18Co cells were maintained in Dulbecco’s modified Eagle’s medium supplemented with 10% fetal bovine serum, 100 U/mL penicillin, and 100 μg/mL streptomycin. All cells were maintained at 37°C in a 5% CO2 humidified incubator. The siRNA was transfected using Lipofectamine 2000 reagent (Invitrogen). Culturing Colon Cancer and Normal Fibroblast Cells

The HT29 colon cancer cell line was purchased from the American Type Culture Collection (Manassas, VA, USA) and the CCD-18Co normal colon fibroblast cell line was purchased from the Korean Cell Line Bank (Seoul, Korea). The colon cancer cell line was cultured in Dulbecco’s modified Eagle’s medium (DMEM; HyClone, Logan, UT, USA) and the CCD18-Co cell line was cultured in RPMI-1640 medium (WelGENE, Daegu, Korea) supplemented with 10% heat-inactivated fetal bovine serum (HyClone) at 37°C in a 5% CO2 incubator.
