Silencing ZFAS1 gene in cell lines

Plasmid extraction kit was obtained from Sangon Biotech (Shanghai, China). The short hairpin RNA (shRNA) for silencing ZFAS1 (shZFAS1#1, shZFAS1#2) and the negative control (shNC) were purchased from Genepharma (Shanghai, China). The pcDNA-ZFAS1, pcDNA-DDX21 and blank vector (NC) were synthesized by Genewiz (Suzhou, China). All of the shRNA nucleotide sequences were listed in Supplementary Tables 3, 4. Cells were cultured on 6-well plates to 60%-70% density and then transfected by using Lipofectamine 2000 (Invitrogen, USA) according to the manufacturer’s instructions. The cells were used for further experiments after 48 hours of transfection.

Cells were collected 48 h after transfection and used for further experiments.
