Gut Microbiome Restoration in Piglets

Ampicillin (1 mg/ml) (A8180, Solarbio, China), streptomycin (1 mg/ml) (S8290, Solarbio, China), vancomycin (0.25 mg/ml) (V8050, Solarbio, China), and neomycin (1 mg/ml) (N8090, Solarbio, China) were added to the milk and fed for 5 days to eliminate as much as possible the gut microbial background of the piglets, followed by FMT or feeding of bacteria or metabolites. Feces were collected on day 8 from the uninfected group of Min pigs and Landrace pigs and treated as described previously. Briefly, fecal samples were placed in a sterile N2 incubator and mixed thoroughly. Then, cell filters were inserted to collect the bacteria and centrifuged at 600×g for 10 min. The samples were then added to 10% glycerol and stored at −80 °C [23 (link)]. Prior to FMT, PBS was used instead of glycerol, and the number of viable bacteria was subsequently counted by methylene blue staining. Two milliliters of the bacterial solution were mixed into the milk and fed to the piglets for three consecutive days, and the PEDV challenge occurred on day 0 by gavage.
