Immunostaining of Brain Sections for Somatostatin

Brain sections were immunostained for somatostatin (Santa Cruz Biotechnology, Santa Cruz, CA; catalog number SC7819) as described in Spiegel et al. (2013) (link). Briefly, free-floating sections were washed in 0.1 M phosphate-buffered saline (PBS), and then endogenous peroxidases were quenched in 0.3% hydrogen peroxide in PBS. After additional PBS washes, sections were blocked in 5% normal horse serum in PBS with 0.3% Triton. Sections were then incubated with primary antibody at a dilution of 1:1,600 in PBS containing 0.15% Triton and 3% normal serum for 72 hr at 4°C with agitation. Following primary antibody incubation, sections were washed in PBS and reacted with biotinylated secondary antibody horse antigoat IgG (Vector Laboratories, Burlingame, CA) diluted in PBS with 0.15% Triton and 5% normal horse serum for 45 min. The secondary antibody was detected with avidin-biotin complex (ABC Elite; Vector Laboratories) and the avidin-biotin complex was visualized with nickel-enhanced diaminobenzadine (Vector Laboratories). Tissue sections were mounted onto gelatin coated slides and dried, dehydrated with increasing ethanol concentrations, cleared in xylene, and coverslipped using DPX mounting media.

Immunohistochemistry of Neuronal Markers

Tissue sections were rinsed in PBS-Triton 0.3% (PBS-T) then blocked in PBS-T with 10% normal donkey serum (NDS) followed by overnight incubation at 4°C in PBS-T-10% NDS with primary antibodies raised against: Calbindin (1:1000, Swant, Marly, Switzerland, CB300 or CB38), Calretinin (1:1000, Millipore Sigma, AB1550), Gad65-67 (1:200 Santa Cruz Biotechnology, sc-365180), GFAP (1:1000, Agilent Dako, Glostrup, Denmark, Z0334), Neuropeptide Y (1:500, Immunostar, Hudson, WI, United States, 22940) or Somatostatin (1:300, Santa Cruz Biotechnology, sc7819). For secondary detection, appropriately matched Alexa Fluor-conjugated secondary antibodies (1:500, Thermo Fisher) were incubated 90 min in PBS-T at room temperature. Sections were incubated with DAPI, mounted in Fluoromount G (Thermo Fisher) and coverslipped before epifluorescence examination with a slide scanner (Leica DMI6000 B, Leica, Wetzlar, Germany).

Immunostaining of Pancreatic Tissues

Pancreatic tissues were fixed in 4% paraformaldehyde and embedded in paraffin. For IHC, 5-μm sections were stained with antibodies or control IgG using the EnVision+System-HRP (DAB) kit (Dako North America Inc, USA) according to the manufacturer’s instructions. For IF, tissues were incubated with antibodies against insulin (Abcam; Santa Cruz Biotechnology), glucagon (Santa Cruz Biotechnology), somatostatin (Santa Cruz Biotechnology), Zhx2 (Proteintech), PAX6 (Santa Cruz Biotechnology), and Ki67 (Abcam) overnight at 4°C and incubated with antibodies against Alexa Fluor 594 goat anti-guinea pig IgG (Abcam), Alexa Fluor 594 goat anti-mouse IgG (Proteintech), and Alexa Fluor 488 goat anti-rabbit IgG (Proteintech) for 1 h at 37°C. Microscopy analysis was performed with an Olympus IX51 microscope and ImageJ software.
