CDK Kinase Assay and Molecular Docking of Natural Products

The ADP-Glo kinase assay kit and CDK enzymes
were purchased from Promega Corporation. Molecular biology-grade dimethyl
sulfoxide (DMSO), deionized water, and Tris buffer were purchased
from Sigma-Aldrich. The group’s NP library (n = 101) used herein is a collection of pure NPs isolated by our group
under several projects, along with a few NPs commercially procured
from different vendors. The source of top actives/hits is provided
here. Rohitukine,28 (link) boeravinone B,29 (link) pongol and structural analogues,30 (link) and helminthosporin20 (link) were obtained from our phytochemistry research. Other NPs, viz.,
rutin (TCI, R0035), chrysin (TCI, C1652), bergenin (TCI, B4349), morin
(TCI, P0041), quercetin (TCI, P0042), baicalein (TCI, T2721), and
fisetin (TCI, T0121), were sourced from commercial vendors. Luminescence
readings in the CDK kinase assay were recorded using a Tecan Infinite
M Plex multimode reader, and the dose–response curve was generated
by using GraphPad Prism. Molecular docking was carried out using Schrodinger
molecular modeling software.
