Western Blotting for Protein Analysis

Proteins were loaded onto a 10% SDS-PAGE gel and separated using Tris-glycine SDS-PAGE running buffer. PageRuler Plus pre-stained protein ladder (26619, ThermoFisher Scientific) was used to indicate the molecular weight of target proteins. After electrophoresis, proteins were transferred to 0.45 μM nitrocellulose membranes (1620115, BioRad) with Towbin transfer buffer. Membranes were blocked with 5% bovine serum albumin and then incubated with corresponding primary antibodies. Membranes were then washed with Tris-buffered saline with 0.5% Tween 20 and incubated with IRDye secondary antibodies. Membranes were scanned and images were collected on an Odyssey CLX (LICOR) imaging system. Membranes were stripped with ReBlot Plus strong antibody stripping solution (2504, Millipore) for the detection of other targets, if necessary. Anti-IRS1 (2390), anti-IRS2 (3089), and anti-PTEN (9559) antibodies were purchased from Cell Signaling Technology. Anti-β-actin (A5441) and anti-Vinculin (V9131) antibodies were purchased from Sigma-Aldrich.
