Nox2 mice

Manufactured by Jackson ImmunoResearch
Sourced in United States

NOX2−/− mice are genetically engineered mice that lack the NOX2 gene, which encodes the NOX2 protein. NOX2 is a critical component of the NADPH oxidase complex, which is responsible for the production of reactive oxygen species (ROS) in various cell types. These mice are a valuable tool for studying the role of NOX2-derived ROS in biological processes and disease models.

Several types of adult mice aged from 10 to 12 weeks were used,
including: WT C57b/6J (Jackson Lab), WT C57b/6N (Jackson Lab), Ser280Ala
KI,25 ,31 (link) MMVV-KI,32 (link) CaMKIIδ cardiac-specific and
global KO,33 (link),34 (link) NOX2−/− mice
(Jackson Lab) and Grx1-roGFP2 (targeted either to the cytosol or
mitochondria).27 (link) All
the genetically modified mouse lines were bred on the C57b/6J background, except
for the C57b/6N line. See Online Methods for more details.
Cardiac ventricular myocytes were isolated using previously described
methods35 (link) which were
approved by the UC Davis Institutional Animal Care and Use Committee (IACUC).
Freshly isolated myocytes were plated on laminin-coated glass cover slips for 30
min before dye loading. All experiments were performed at room temperature (RT)
(22–23°C) with pH 7.4.

Timed Pregnancy Rodent Models

Check if the same lab product or an alternative is used in the 5 most similar protocols
SD rats were from Peking Vital River Laboratory and Centre of Experimental Animal of Peking University. C57 BL/6J (NOX2 wild type, NOX2+/+) and NOX2−/− mice were from Jackson Laboratories (Bar Harbor, Maine, USA). Breeding schedules were designed to achieve accurately timed-pregnancy of 14 ± 0.5 days for rats and 13 ± 0.5 days for mice. Housing, breeding and experimental use of the animals were performed in strict accordance with the National Institutes of Health guidelines.

Evaluating NOX2 Deficiency in Mice

Check if the same lab product or an alternative is used in the 5 most similar protocols
All studies were approved by the Cincinnati Children's Hospital Medical Center's Institutional Animal Care and Use Committee. All mice used in this study were on a C57BL/6 background, housed in the same room within a specific pathogen‐free facility but not as littermates, fed food and water ad libitum, and provided care in accordance with the Guide for the Care and Use of Laboratory Animals. Wild‐type (WT) mice (strain name C57BL/6J) and NOX2−/− mice (strain name B6.129S‐CybbtmDin/J) were obtained from Jackson Laboratories and were subsequently bred at the Cincinnati Children's Hospital Medical Center.

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
