Western Blot and qRT-PCR Analysis of Tumor Proteins

Proteins within tumour tissues and HUVEC cells were extracted using a Boster Kit (Bosterbio, CA, USA) according to the manufacturer’s instructions. Western blotting was performed as previously described [20 (link)]. The antibody, which was raised against the Cyp2c44’s IGRHQPPSMKDKMKC peptide (GenScript), was generated according to previous studies [13 (link), 16 (link)]. Other antibodies used in this study are as follows: Glyceraldehyde 3-phosphate dehydrogenase (GAPDH) (Bosterbio), β-actin (Bosterbio), Cyp2c9 (Abcam, Cambridge, UK), COX1 (Santa Cruz, CA, USA), COX2 (Santa Cruz), Phosphorylated extracellular signal-regulated kinase 1/2 (p-ERK1/2) (Santa Cruz), ERK1/2 (Santa Cruz), P-AKT (Abcam), AKT (Abcam), and CD31 (Abcam).
For qRT-PCR, RNA from tumours was extracted using TRIzol (Invitrogen, Carlsbad, CA, USA) according to the manufacturer’s instructions and reverse-transcribed using the M-MLV First-Strand cDNA Synthesis Kit (Invitrogen) [15 (link)]. The mRNA levels of target genes were quantified by qRT-PCR using Power SYBR Green PCR Master Mix (Invitrogen) with the primers listed in Additional file 1: Table S1. GAPDH served as an internal control and the results were analysed using the 2-ΔΔCt method.

Ltd. (Wuhan, China). The HRP-conjugated secondary antibodies (Cat# bs-40295G-HRP) were bought from Bioss, Inc. (Beijing, China). The lactate dehydrogenase (LDH) kit, creatine kinase-MB (CK-MB) kit, α-hydroxybutyrate dehydrogenase (α-HBDH) kit, free-fatty acid (FFA) kit, and enzyme-linked immunosorbent assay (ELISA) kits for rat interleukin-6 (IL-6), nuclear factor kappa-B (NF-κB), and tumor necrosis factor-alpha (TNF-α) were purchased from Jiancheng Bioengineering Institute (Nanjing, China). All other chemicals and solvents applied were of analytical grade.

Streptozotocin-Induced Diabetes Model in Rats

The Animal Ethics Committee of University of South China (Hengyang, China) has approved this experimental protocol. Forty male adult SD rats (weighing 280 ± 20 g), which were purchased from the Animal Experimental Center of University of South China (Hengyang, China) and Animal qualified number is SYXK (hunan) 2015-0006,were fed in a clean laboratory with constant temperature (22 ± 1 C) and artificial lighting (day and night 12 h each). Food and water were free to all rats. Streptozotocin (STZ) was provided by MP Biomedicals, LLC (USA), Sodium hydrosulfide (NaHS) was purchased from Sigma-Aldrich LLC (USA). Rabbit polyclonal antibodies against collagen I, collagen III, TGF- β 1, CTGF were purchased from Bioss Biology Company (Beijing, China), rabbit polyclonal antibodies against MMP11, TIMP2, NF- κ Bp65, TNF- α , PPARG, HSP70, HSP90, PKC α , glyceraldehyde 3-phosphate dehydrogenase (GAPDH) and Anti-rabbit secondary antibody were all provided by Boster Biotechnology (Wuhan, China), and rabbit polyclonal antibody against ERK1/2 was obtained from Cell Signaling Technology (USA). Besides, bicinchoninic acid (BCA) Protein Assay Kit and cell lysis buffer were obtained from Beyotime Company (Shanghai, China).

Protein Expression Analysis in PC-14/B Cells

Total cellular protein was extracted from PC-14/B cells at 72 h post-infection using lysis buffer (Beyotime Institute of Biotechnology, Shanghai, China), followed by the Western blotting analysis of SIRT1, P53, and KAI1(CD82), as well as matrix metalloproteinase (MMP)-9, E-cadherin, and β-catenin using the standard methods. In brief, protein samples were separated by 10% sodium dodecyl sulfate-polyacrylamide gel electrophoresis and electrotransferred onto polyvinylidene fluoride membranes (Millipore). Primary incubation was performed using specific antibodies (1:200-600) at 4°C for 12 h. All antibodies were purchased from Cell Signaling Technology (Danvers, MA, USA). Glyceraldehyde 3-phosphate dehydrogenase (GAPDH; 1:1200, Boster Biotechnology, Wuhan, China) was used as the reference protein. Horseradish peroxidase-conjugated goat anti-rabbit/rat immunoglobulin G secondary antibodies (1:20,000, Boster Biotechnology) were used for secondary incubation. Image-Pro Plus 6.0 software (Media Cybernetics Inc., Bethesda, MD, USA) was used for the digital analysis.
