Polymer Nanoparticle Synthesis and Characterization

The following chemicals were obtained from commercial suppliers and used as received: the polymer poly(d,l-lactic-co-glycolic acid) 50 : 50 (PLGA; MW 38 000–54 000 Da, Tg 46–50 °C), under the commercial name of Resomer® RG 504 was purchased from Boehringer Ingelheim (Ingelheim, Germany) and Evonik Industries (Evonik Röhm GmbH, Germany). Sodium cholate, chloroauric acid (HAuCl4·3H2O), sodium citrate tribasic dehydrate ≥99.0% (Na3C6H5O7·2H2O), PVA (polyvinyl alcohol) MW 85 000–124 000 Da, 97–99% hydrolyzed, taurocholic acid sodium salt hydrate ≥95% (TLC), ethyl acetate ACS reagent, dodecanethiol, phosphotungstic acid hydrate as a contrast agent for microscopy and fluorescent molecule pyrene ≥99.0% were purchased from Sigma Aldrich (St Louis, MO, USA).

Biodegradable PLGA Polymer Formulations

Poly(D,L-lactide-co-glycolide) (PLGA) 50:50 Resomer ® RG 504 (Mw 40-50 kDa; ester terminated), poly(D,L-lactide) Resomer ® R 202 H (Mw 10-18 kDa; acid terminated) and poly(D,L-lactide)
Resomer ® R 202 S (Mw 10-18 kDa; ester terminated) were supplied by Boehringer Ingelheim (Ingelheim, Germany). Prior to use, the polymer was dried under vacuum for 24 h to eliminate traces of moisture. Ketoprofen (KET) was kindly provided by Bidachem SpA (Bergamo, Italy), while methylene chloride (CH2Cl2) was supplied by Sigma-Aldrich (Milan, Italy).

Encapsulation of Uncaria tomentosa Extract

Uncaria tomentosa dried extract (4.5% of total pentacyclic oxindole alkaloids) was kindly donated by Herbarium Laboratório Botânico (PR, Brazil). Poly-e-caprolactone (PCL; Mw 70,000-90,0000 g/mol), polyvinyl alcohol (PVA; Mw 31,000-50,000 g/mol, 87-89% hydrolyzed) and cellulose dialysis membranes (flexible tube type, 33 mm, 14 kDa MWCO) were obtained from Sigma-Aldrich (MO, USA). Poly-d,l-lactide-coglycolide (PLGA), with 50:50 D,L-lactide:glycolide molar ratio and intrinsic viscosity of 0.45-0.60 dl/g (Resomer® RG 504), was purchased from Boehringer Ingelheim (RP, Germany) and poloxamer 188 (Lutrol® F68, Mw 7680-9510 Da) from BSAF (RP, Germany). Methylene chloride, ethyl acetate, acetone and ethanol were acquired from Tedia Brazil (RJ, Brazil).
