Topical Clotrimazole Formulation Development

Clotrimazole and cholesterol were purchased from Alfa Aesar (Ward Hill, MA, USA). Sodium oleate was purchased from TCI America (Portland, OR, USA). Methanol (ACS grade), methanol (HPLC grade), PBS (pH 7.4), and chloroform were procured from Fisher Chemicals (Fair Lawn, NJ, USA). Hydroxy propyl methyl cellulose (HPMC) (MW: 86,000, viscosity 4000 cP at 2% solution) was obtained from Acros Organics (Fair Lawn, NJ, USA). Cadaver skin for permeation and skin diffusion studies was obtained from Zen-Bio Inc., (Research Triangle Park, NC, USA). Marketed Clotrimazole 1% cream (Perrigo) was purchased from local pharmacy store in El Paso, TX, USA. Deionized water (resistivity of 18.2 MΩ) used for all experiments was obtained from in-house Milli-Q® IQ 7000 Ultrapure Water System (EMD Millipore, Bedford, MA, USA).

Repositioning Compound Library Evaluation

The Pharmakon 1600 repositioning compound library was purchased from
MicroSource Discovery Systems, Inc. (Gaylordsville, CT) and the Johns Hopkins
library was provided by Johns Hopkins University (Baltimore, MD). Both libraries
were supplied in 96-well plates of 10 mM stocks of the compounds in either
dimethyl sulfoxide or water and stored in −80° C. Ornidazole,
miconazole nitrate, econazole nitrate, tioconazole, butoconazole, clotrimazole,
metoprolol tartrate, metoclopramide hydrochloride, chloroquine diphosphate,
miconazole, dimetridazole, nithiamide and methylthiouracil (Alfa Aesar, Ward
Hill, MA), dichlorophen, triclabendazole, closantel, nitazoxanide (Ark Pharm
Inc, Libertyville, IL), ronidazole, oxyclozanide (Sigma-Aldrich, St. Louis, MO),
tinidazole (TCI, Portland, OR) and niclosamide (Cayman Chemical, Ann Arbor, MI)
were all purchased separately to confirm the results of both libraries.
Vancomycin hydrochloride (Gold Biotechnology, Olivette, MO) and metronidazole
(BTC, Hudson, NH) were used as positive controls.

Cabazitaxel Solubility Enhancement

The following materials were obtained: Cabazitaxel (Carbosynth Cat. # FC19621); deuterated cabazitaxel (1 mg reference standard; Toronto Research Chemicals Cat. # C046502–1MG); Pluronic F127 (Sigma Cat. # P2443); clotrimazole (Alfa Aesar Cat. # J63895), ketoconazole (Alfa Aesar Cat. # J63367), cobalt nitrate hexahydrate (Alfa Aesar Cat. # 36418), ammonium thiocyanate (Alfa Aesar Cat. # A10632), Tween-80 / Polysorbate 80 (VWR Cat. # EM-9490), nimodipine (TCI Cat. # N0896), and mifepristone (TCI Cat. # M1732). The following solvents were used: ethyl acetate (Fisher), acetone (Fisher), methylene chloride (Fisher), acetonitrile (Fisher), and ethanol (Decon).

Yeast Strain Generation by Homologous Recombination

Deletion and GFP-fusion strains were obtained by homologous recombination of the PCR product with heterologous selection markers, as listed in Table 1. All strains in this study are derivatives of the W303 strain. As templates for PCR, the DNA of yeast strains from Euroscarf collection, pUG27 (Gueldener et al., 2002 (link)) and pFA6a-His3MX6 (Longtine et al., 1998 (link)) plasmids were used. All newly generated strains were verified by PCR with independently designed primers (Table 2) and RT-qPCR (Supplementary Text S1, Table S1, Figure S1). We used standard yeast-rich and synthetic mediums described by Sherman (2002) (link). We obtained yeast extract from BD and D-glucose from Helicon. Clotrimazole, miconazole, nigericin, amphotericin B, propanol, butanol, and FM4–64 were obtained from Thermo Fisher Scientific. Bacto Agar, peptone, NaCl, KCl, and NaN3 were obtained from Amresco, and 2-Deoxy-D-glucose was obtained from Chem-Impex Int’l Inc.

Synthesis and Characterization of Glycidyl Ethers

1,1,1-Tris(hydroxymethyl)propane
(Sigma-Aldrich) was dissolved in acetone and precipitated by addition
of diethyl ether. Then, 60 wt % NaH in mineral oil (Merck) was washed
with dry tetrahydrofuran (THF) to remove oil and dried. The oil was
removed by washing with dry tetrahydrofuran, and then drying under
reduced pressure. Benzyl glycidyl ether (BGE, Sigma-Aldrich) was dried
over CaH2 and then distilled under reduced pressure before
use.
d,l-1,2-Isopropylidene glyceryl glycidyl
ether (IGG) was synthesized according to the procedure described by
Wurm et al.33 (link) THF (Sigma-Aldrich) was dried
over a Na/K alloy. Dimethyl sulfoxide (DMSO, Sigma-Aldrich) was purified
and dried according to the procedure described in ref (34 ) and stored over 4 Å
molecular sieves.
Nifuratel (TCI) and clotrimazole (Thermoscientific)
were used as
received without any further purification. A Strat-M membrane was
used as a nonanimal model for transdermal diffusion testing to predict
the diffusion of drugs through human skin.
The simulated vaginal
fluid (SVF) of pH = 4.2 was prepared according
to the composition described in ref (35 (link)).
