Kinetics of H2O2 release from cells

The kinetics of H 2 O 2 released from cells was done using Amplex® Red (10-acetyl-3, 7-dihydroxyphenoxazine) hydrogen peroxide/peroxidase assay kit (Thermo Fisher Scientific, Waltham, MA, USA) as per the kit protocol with minor modifications. Briefly, spent medium was removed from the wells of 96-well culture plate after 72 hours of siRNA treatment and Krebs-Ringer phosphate (KRPG) buffer added to the cells. This was followed by 100µL of reaction mixture containing 50µM Amplex® Red reagent and 0.1U/mL HRP in KRPG. Amplex® Red reagent reacts with H 2 O 2 to produce resorufin. Fluorescence (excitation at 530-560nm and emission detection at 590nm) was measured immediately, and subsequently measured every 30 minutes until 90 minutes on Tecan Infinite 200 (Tecan, Männedorf, Switzerland).
