VEGF Quantification in Heart Tissue

VEGF expression was measured using the Quantikine Human VEGF Immunoassay ELISA kit (R&D Systems, Inc. Minneapolis, MN 55413). This is a solid phase ELISA designed to measure VEGF165 levels in the supernatant of the heart. It contains Sf 21-expressed recombinant human VEGF165 and antibodies raised against the recombinant protein. The kit includes plates pre-coated with a monoclonal antibody specific for VEGF. Tissue samples were homogenized and standards and samples were pipetted into the wells. Any VEGF present was bound by the immobilized antibody. After washing away any unbound substances, an enzyme-linked polyclonal antibody specific for VEGF was added to the wells. Following a wash to remove any unbound antibody-enzyme reagent, a substrate solution was added to the wells, and color developed in proportion to the amount of VEGF bound in the initial step. The color development was stopped and the intensity of the color was measured.
