RNA oligonucleotides were purchased from Biomers.net (Ulm/Donau, Germany). A 20-nucleotide (nt) template sequence (T
20) corresponding to the 3′ end of the DENV2 antigenome was annealed to a 10-nt complementary primer (P
10) containing a fluorescent 6-carboxyfluorescein (FAM) moiety at the 5′ end. Annealing was performed using a primer-template molar ratio of 1:1.5 in the presence of 110 mM KCl, incubated at 70°C for 10 min, and cooled slowly to room temperature. NTPs were purchased from GE Healthcare (Chicago, IL).
