RNA Oligonucleotide Preparation and Annealing

Check if the same lab product or an alternative is used in the 5 most similar protocols
RNA oligonucleotides were purchased from Biomers.net GmbH (Ulm, Germany). The sequences of the substrates used are:
ssRNA 38 mer
5′-AUGAAGGUUUGAGUUGAGUGGAGAUAGUGGAGGGUAGU-3′
ssRNA 18 mer FAM
3′-UACUUCCAAACUCAACUC-5′ FAM
For dsRNA, oligonuclotides were mixed at 1:1 (M/M) ratio in an annealing buffer (30 mM HEPES–KOH pH 7.4, 100 mM KCl, 2 mM MgCl2, 50 mM NH4Ac) at a final concentration of 500 nM, heated at 95 °C for 5 min and then slowly cooled down at room temperature.

DENV2 Antigenome Sequence Analysis

Check if the same lab product or an alternative is used in the 5 most similar protocols
RNA oligonucleotides were purchased from Biomers.net (Ulm/Donau, Germany). A 20-nucleotide (nt) template sequence (T20) corresponding to the 3′ end of the DENV2 antigenome was annealed to a 10-nt complementary primer (P10) containing a fluorescent 6-carboxyfluorescein (FAM) moiety at the 5′ end. Annealing was performed using a primer-template molar ratio of 1:1.5 in the presence of 110 mM KCl, incubated at 70°C for 10 min, and cooled slowly to room temperature. NTPs were purchased from GE Healthcare (Chicago, IL).

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
