Generation and Validation of THP-1 Cell Lines

Check if the same lab product or an alternative is used in the 5 most similar protocols
THP-1 human monocyte-like cells (obtained from ATCC) were cultivated in RPMI with 10% FBS and 50 µM 2-mercaptoethanol. THP-1 cell lines inducibly expressing HA-tagged EGFP, ICEBERG, and INCA were generated using lentivirus produced with derivatives of pInducer2014 and selected in the presence of 500 µg/ml G418. Monoclonal cell lines were obtained by limited dilution in THP-1 medium with 20% FBS. For testing mycoplasma contamination, wild-type and derived THP-1 cells were stained with Hoechst 33342 and analyzed at high magnification. No punctate DNA staining was observed in the cytosol. Alexa Fluor 488 conjugated anti-HA mouse IgG monoclonal antibody (clone 16B12) from Life Technologies (Cat#A21287) was used to confirm HA-ICEBERG and HA-INCA expression in modified THP-1 cells.

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
