Alexa fluor 700 antihuman ki67 antibody

Manufactured by BioLegend

The Alexa Fluor 700 antihuman Ki67 antibody is a fluorescently labeled antibody that specifically binds to the Ki67 protein, which is a marker of cell proliferation. This antibody can be used in flow cytometry and other immunoassays to detect and quantify Ki67-expressing cells.

Cryopreserved peripheral blood mononuclear cell (PBMC) samples from baseline and post-treatment (cycles 1–12) were thawed and rested overnight at 37°C. Then cells were stained with a viability dye (eBioscience Fixable Viability Dye eFluor 450) and a master mix of antibodies for surface stains including CD4-BV605 (BioLegend Cat# 317438, RRID:AB_11218995), CD3-PE-Cyanine5.5 (Invitrogen Cat# 35-0036-42, RRID: AB_11220085), CD8a-PE-Cyanine7 (Invitrogen Cat# 25-0088-42, RRID: AB_1659702), PD1-APC (BioLegend Cat# 329908, RRID: AB_940475), CD152-PE-Cy5 (BD Biosciences Cat# 555854, RRID:AB_396177). Cells were next fixed and permeabilized with the eBioscience Foxp3/Transcription Factor Staining Buffer Set (ThermoFisher) and subsequently stained intracellularly with Alexa Fluor 700 antihuman Ki67 antibody (BioLegend Cat# 350530, RRID: AB_2564040). Stained cells were acquired on a BD Canto RUO and analyzed with FlowJo software (FlowJo, RRID:SCR_008520).

Quantifying Luciferase-Positive Melanoma Cells

Check if the same lab product or an alternative is used in the 5 most similar protocols
Mouse lung tissues were dissociated into single cell suspensions using Lung Dissociation Kit (Miltenyi Biotec, #130-095-027) according to the manufacturer’s instructions. The cells were fixed and permeabilized with cold 70% ethanol, washed with PBS twice, and stained with fluorochrome-conjugated antibodies for 1 hr at RT in dark condition. Alexa Fluor 647 anti-firefly luciferase antibody was purchased from Abcam (Cambridge, MA), Alexa Fluor 700 anti-human Ki-67 antibody was purchased from Biolegend (San Diego, CA), and Alexa Fluor 488 anti-human p27 antibody was purchased from Santa Cruz (Dallas, TX). Each Ki-67 and p27 fluorescence signal was gated and analyzed the signal intensity from the first gating of melanoma cells with high fluorescence intensity of Alexa Fluor 647 luciferase signal. Flow cytometry was performed with BD LSR Fortessa™ Flow Cytometer (BD Biosciences) and data was analyzed with FlowJo (FlowJo Inc.).

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
