Pesticide Residue Analysis in Rape Flowers

A Milli-Q ultrapure water system manufactured by Millipore (Milford, UT, USA) was used throughout experiments to provide the HPLC-grade water that was employed during analysis and to hydrate the rape flower samples. Ultragradient HPLC-grade acetonitrile and methanol were obtained from Tedia (Shanghai, China). Formic acid, acetic acid, anhydrous magnesium sulfate (MgSO4), and sodium acetate (NaOAc) were supplied by Sinopharm Chemical Reagent Co., Ltd. (Shanghai, China). PSA (primary and secondary amine-bonded silica), C18 (octadecyl-bonded silica) and GCB (graphitized carbon black) were purchased from Agilent Technologies (Santa Clara, CA, USA).
Reference standards of imidacloprid (purity, 98%), thiamethoxam (purity, 95%), acetamiprid (purity, 95%), fenpropathrin (purity, 98.5%), chlorpyrifos (purity, 98%), carbendazim (purity, 95%), prochloraz (purity, 98%) and pyrimethanil (purity, 95%) were obtained from Dr. Ehrenstorfer GmbH (Augsburg, Germany).
Formulated pesticides of imidacloprid 10% wettable powder (WP), thiamethoxam 25% water-dispersible granules (WDG), acetamiprid 20% WP, fenpropathrin 20% emulsifiable concentrate (EC), chlorpyrifos 40% EC, carbendazim 80% WP, prochloraz 25% EC, and pyrimethanil 40% suspension concentrate (SC) were purchased from the agricultural market located in Hefei, Anhui, China.

Comprehensive Pesticide Residue Analysis

All the organic solvents used were higher performance liquid chromatography (HPLC) grade.

Pesticide Exposure Effects on Honey Bees

Technical grade acetamiprid (purity 99.7%) and carbaryl (purity 98.9%) were purchased from Dr. Ehrenstorfer GmbH Corporation, Germany. Stock solution of acetamiprid or carbaryl was prepared by dissolving the powder in acetone and then diluted with diet C (50% royal jelly, 2% yeast extract, 9% d-glucose, 9% d-fructose) as described in Schmehl et al.72 (link). The final concentration of 2 mg/L carbaryl or 5 mg/L acetamiprid were applied to the third instar larvae for 4 days. The concentrations respectively correspond to the NOAEC determined in our previous study27 (link). The solvent solution for control groups was identical but pesticide free (The solvent accounted for 0.5% of the final diet). Samples were collected for enzyme activity assay or immediately frozen in liquid nitrogen and stored at − 80 °C until RNA and metabolites extraction.

Neonicotinoid Insecticide Analysis

All reagents were analytical grade or higher. Five standards of neonicotinoid insecticide including clothianidin, imidacloprid, acetamiprid, and thiamethoxam were obtained from Dr. Ehren-storfer GmbH (Augsburg, Germany), and thiacloprid were purchased from Sigma-Aldrich (Taufkirchen, Germany). All standard neonicotinoid insecticides were of analytical standard grade. The stock solution of each insecticide (1000 µg mL−1) was prepared by dissolving an appropriate amount in methanol. Working standard solutions were prepared by diluting the stock standard solution with water. Chromatographic grade methanol and acetonitrile were purchased from Merck (Darmstadt, Germany). Sodium chloride (NaCl), anhydrous sodium sulphate (anh. Na2SO4), sodium carbonate (Na2CO3) and sodium hydroxide (NaOH) were obtained from Ajax Finechem (Auckland, New Zealand), Sodium acetate (CH3COONa) was obtained from Carlo Erba (Val de Reuil, France). Triton™ X-114 was obtained from Sigma-Aldrich (St. Louis, MI, USA). Sodium dodecyl sulfate (SDS) and Triton™ X-100 were provided by Merck. Cetyltrimethyl ammonium bromide (Calbiochem, Germany) was also used. Water used throughout the experiment was produced by RiOsTM Type I Simplicity 185 (Millipore Waters, Burlington, MA, USA) with the resistivity of 18.2 MΩ.cm.

Anti-Imidacloprid Monoclonal Antibody Production

Nickel nitrate 6-hydrate and zinc nitrate 6-hydrate were obtained from Elago Enterprlses pty (New South Wales, Australia). Sodium hydroxide (NaOH) was obtained from Ajax Finechem (New South Wales, Australia). Formic acid was provided by JT Baker (Phillipsburg, NJ, United States).

Anhydrous sodium sulfate (Na 2 SO 4 ), primary-secondary amine (PSA) bonded silica bulk, octadecylsilane (LC-C18, 40-63 μm) and nylon syringe filters (0.22 μm) were obtained from Shanghai Anpel Scientific Instrument Co., Ltd. (Shanghai, China). Ultra-pure water was obtained from a Milli-Q system (Bedford, MA, USA).
