LC/MS-based Lipidomic Analysis Workflow

Liquid chromatography/mass spectrometry (LC/MS)-grade methanol, acetonitrile, 2-propanol, ammonium formate, and HPLC-grade methyl tert-butyl ether (MTBE) were obtained from Fisher Scientific (PA, USA). LC/MS-grade ammonium formate was from Sigma-Aldrich (MO, USA). A Milli-Q system (MA, USA) was used to prepare ultra-pure water (18.2 MΩ).
LysoPhosphatidylcholine (LPC 19:0), Phosphatidylethanolamine (PE 12:0/13:0), Ceramide (Cer d18:1/17:0), Sphingomyelin (SM (d18:1/12:0), TG (15:0/15:0/15:0), and Phosphatidylcholine (PC 12:0/13:0) were purchased for use as internal standard (IS) compounds from Avanti Polar Lipids (AL, USA). Antibodies used in this study were rabbit anti-acid Sphingomyelinase polyclonal antibody (Absin, Shanghai, China).

Quantitative Bioanalysis of Cyclophosphamide

Cyclophosphamide was purchased from Sigma-Aldrich (St. Louis, MO). 4-hydroperoxy-Cyclophosphamide, which is then reduced in water to the 4-hydroxyCyclophosphamide metabolite, was purchased from Asta Medica (Frankfurt, Germany). Deuterated (2[H4]) Cyclophosphamide (d4-CP) was purchased from Santa Cruz Biotechnology (Dallas, TX), while AZD7451 mesylate was a generous gift from AstraZeneca. All analyte and internal standard structures are presented in Figure 1.
Human whole blood (with EDTA) was provided by the National Institutes of Health Blood Bank. Optima-brand acetonitrile and methanol, and HPLC-grade methyl-tert-butyl ether (MTBE) were purchased from Fisher Scientific (Pittsburgh, PA). Phenylhydrazine, sodium citrate, sodium hydroxide, and formic acid were purchased from Sigma-Aldrich (St. Louis, MO). All water for the assay was deionized and ultra-filtered through a MilliPore system (EMD MilliPore, Billerica, MA).

Quantitative Analysis of Active Compounds

Paeoniflorin (C23H28O11, MW: 480.5, purity >98%) and liquiritin (C21H22O9, MW: 418.4, purity >98%), the main active components of Paeonia lactiflora Pall. and Glycyrrhiza uralensis Fisch. ex DC., respectively, were purchased from Nanjing Yuanzhi Biotechnology Co., Ltd. (Nanjing, China). LC-MS grade methanol, acetonitrile, formic acid, and isopropanol were purchased from Fisher Scientific (Waltham, MA, United States). HPLC grade methyl tert butyl ether (MTBE) was obtained from Fisher Scientific (Waltham, MA, United States). The standard lipids were obtained from Avanti Polar Lipids (United States). Deionized water was purified using a Milli-Q system (Millipore, Bedford, MA, United States). All other chemicals and reagents used in this study were of analytical grade.

Optimizing Micronutrient Supplementation for Marine Organisms

The commercial krill and Calanus oil supplements Prowise SUPERBAkrill Oil, Lot 1074600100 (Prowise Healthcare Ltd, London, UK) and Arctic Ruby (Calanus) Oil, Lot 1702071 (Calanus AS, Tromsø, Norway) were purchased online. HPLC grade methyl tert-butyl ether (MTBE) was supplied by Fisher Scientific (Dublin, Ireland).

LC-MS water, methanol (MeOH), acetone, hexanes, hydrochloric acid, formic acid, HPLC grade methyl tert-butyl ether (MtBE), pesticide grade isooctane, sulfuric acid, ammonium hydroxide, and hydrochloric acid were all purchased from Fisher Scientific (U.S.A.).
Ultrapure water was prepared using a MilliQ water purification system purchased from Millipore Corporation (Billerica, MA, USA). Oasis HLB, Oasis WAX and glass Oasis HLB glass cartridges were all purchased from Waters (MA, U.S.A.).
