Cell counting kit 8 cck 8 protocol

Manufactured by Dojindo Laboratories
Sourced in Japan

The Cell Counting Kit-8 (CCK-8) is a colorimetric assay used to determine the number of viable cells in cell proliferation and cytotoxicity assays. It utilizes the highly water-soluble tetrazolium salt WST-8, which is reduced by dehydrogenases in viable cells to produce a yellow-colored formazan dye. The amount of formazan dye generated is proportional to the number of living cells.

Cell Proliferation and Colony Formation Assay

Check if the same lab product or an alternative is used in the 5 most similar protocols
Treated cells (1 × 106 for AGS and SGC7901) were seeded in triplicate into 96-well plates and allowed to incubate for 24 h, 48 h and 72 h. Cell viability was assessed according to the Cell Counting Kit-8 (CCK-8) protocol (Dojindo, Kumamoto, Japan). For colony formation, treated GC cells were seeded in 6-well plates at 200 cells per well 23 . Two weeks later, colonies were fixed with 4% paraformaldehyde and stained with 0.5% (W/W) crystal violet (diluted in phosphate buffer saline, PBS) for 30 min.

Cell Viability Assay with RA16 Aptamer

Check if the same lab product or an alternative is used in the 5 most similar protocols
NCI-H460 and other cells (2 × 104 cells per well) were seeded in 96-well plates. After 24 hr incubation, cells were treated with RA16 or scrambled aptamer (sequence in Table S2) added in fresh medium at different concentrations. Following incubation for 48 hr, cell viability was determined as per the Cell Counting Kit-8 (CCK-8) protocol (Dojindo, Tokyo, Japan). Absorbance was measured at 450-nm wavelength using a microplate reader (Thermo Fisher Scientific, USA).

Cisplatin Cytotoxicity Assay in Gastric Cancer

Check if the same lab product or an alternative is used in the 5 most similar protocols
Stable transfected cells were seeded in 96-well plates at a density of 5 ×103 cells/well and incubated for 24 h. Cell viability was assessed every day or after being treated with cisplatin (25 μM for AGS and MKN28; APExBio, Houston, TX) for 24 h according to the Cell Counting Kit-8 (CCK-8) protocol (Dojindo, Kumamoto, Japan). All experiments were performed at least in triplicate.

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
