Puquitinib Efficacy in Xenograft Tumors

Female nude mice (Balb/cA‐nude, 5–6 week old) were purchased from Shanghai Laboratory Animal Center, Chinese Academy of Sciences (Shanghai, China). Human carcinoma xenografts were established by inoculating nude mice subcutaneously with 5–9 × 106 cells. When tumors reached a volume of 100–300 mm3, mice were randomly assigned to control and treatment groups. Control groups were given vehicle alone, and treatment groups received oral puquitinib or CAL‐101 daily. Daunorubicin or cytarabine was administered intravenously (i.v.), and co‐treatment groups also received oral puquitinib daily. Tumor volume was calculated as (length × width2)/2.
Pharmacokinetic/pharmacodynamic studies were carried out as described previously.17 Mice bearing MV4;11 tumors received a single dose of 60 mg/kg puquitinib or vehicle, and tumor tissues and blood were collected at different times post‐dosing. Concentrations of puquitinib in plasma and tissue were determined by HPLC/tandem mass spectrometry. Tumor samples were analyzed by western blotting. Animal experiments were conducted in accordance with the Institutional Animal Care and Use Committee guidelines of the Shanghai Institute of Materia Medica, Chinese Academy of Sciences.

Pharmacokinetics of TQ05310 in U-87 MG/IDH2-R140Q Xenografts

Female nude mice (Balb/cA‐nude, 5‐6 weeks old) were purchased from Shanghai Laboratory Animal Center, Chinese Academy of Sciences. Tumor models were established by s.c. inoculation of U‐87 MG or U‐87 MG/IDH2‐R140Q cells into nude mice. When tumors reached a volume of 100‐300 mm3, mice bearing U‐87 MG/IDH2‐R140Q tumors received a single oral dose of TQ05310 (18.05 or 54.14 mg/kg), and then tumor tissue and blood were collected at multiple time points (0, 0.5, 2, 6, 12, 24, 48, and 72 hours) post‐dosing. TQ05310 and 2HG levels were analyzed by LC/MS‐MS. All animal experiments were carried out in accordance with guidelines of the Institutional Animal Care and Use Committee of the Shanghai Institute of Materia Medica, Chinese Academy of Sciences.

Xenograft Tumor Growth Inhibition in Nude Mice

Female nude mice (Balb/cA‐nude, 5‐6 weeks old), purchased from Shanghai Laboratory Animal Center (Chinese Academy of Sciences, Shanghai, China), were housed in sterile cages under laminar airflow hoods in a specific pathogen‐free room with a 12‐hour light/12‐hour dark schedule, and fed autoclaved chow and water ad libitum. Human tumor xenografts were established by s.c. inoculating cells into the left axilla of nude mice. When tumor volumes reached 100‐200 mm3, mice were divided randomly into control and treatment groups. Control groups were given vehicle alone, and treatment groups received oral anlotinib or sunitinib daily. Tumor volume was calculated as (length × width2)/2. Tumor growth inhibition was calculated from the start of treatment by comparing changes in tumor volumes for control and treatment groups.

Nude Mice Xenograft Cancer Model

Human tumor xenografts of Bel-7402 and HCT 116 cells were established by subcutaneously inoculating cells into nude mice. When tumor volumes reached 100–200 mm3, mice were randomly assigned to control and treatment groups and treated with vehicle, zardaverine (p.o.) or CPT-11 (i.p.) respectively. Tumor volume was calculated as (length×width2)/2.
