Body Fat Measurement via ECHOMRI

Check if the same lab product or an alternative is used in the 5 most similar protocols
Body fat was measured at 5 and 6 months using ECHOMRI (Echo Medical Systems ECHOMRI™, Houston, TX). For the test, a conscious mouse is placed in a clear cylindrical holder (4.7 cm diameter). The holder is placed in the MRI device and the animal is scanned. Three values were recorded and averaged for each parameter.

Body Composition Analysis by NMR

Check if the same lab product or an alternative is used in the 5 most similar protocols
Body composition was assessed using nuclear magnetic resonance (NMR) measurements (EchoMRI, Echo Medical Systems, Houston, TX, USA), and EchoMRI software (version 2007.08.10) at baseline (12 weeks of age) and following 16 weeks of HFD. The instrument was calibrated prior to each measurement session, using a special phantom provided by the manufacturer. NMR measurements were made by placing live mice into a thin wall plastic cylinder (4.7 cm, inside diameter; 0.15 cm thick), with freedom to turn about but limited to ∼4-cm vertical movements by a plastic insert. Relative lean mass and fat mass values were calculated by dividing absolute values by body weight.

Body Composition Analysis Protocol

Check if the same lab product or an alternative is used in the 5 most similar protocols
Body composition to determine lean mass and fat mass values were obtained prior to the experiment with Echo MRI (Echo Medical Systems, Houston, Texas) using a 3-in-1 Echo MRI Composition Analyzer. No significant differences in body mass or composition were observed for this experiment between groups.

Body Weight and Fat Mass Dynamics

Check if the same lab product or an alternative is used in the 5 most similar protocols
The body weight was measured every 6 days from day 27 until day 45.
By the end of the injection schedule, MRI was performed with rats placed in a clear plastic holder without sedation and inserted into an Echo MRITM device (Echo Medical Systems, Houston, TX, USA) for measurement of the body fat mass composition.

Comprehensive Metabolic Assessment of Mice

Check if the same lab product or an alternative is used in the 5 most similar protocols
Body composition was measured by MRI using an EchoMRI (Echo Medical Systems). Indirect calorimetry was performed in groups of 7–8 mice using a Comprehensive Laboratory Animal Monitoring System (Columbus Instruments) with free access to food and tap water. Mice were individually housed at room temperature, and a standard 12-hour light/dark cycle was maintained throughout the measurements. Mice were acclimated to the cages for a period of 48 hours before the start of 4 days of measurements at 20-minute intervals. Food intake was assessed by real-time feed weight measurements. Oxygen consumption and carbon dioxide production were measured, and based on this respirometry, EE as well as CHO and FA oxidation were calculated as previously described (82 (link)).

Metabolic Profiling of Mice Using Echo-MRI

Check if the same lab product or an alternative is used in the 5 most similar protocols
Body composition was measured with Echo-MRI (Echo Medical Systems). After body composition measure mice were individually placed in the Oxymax Comprehensive Lab Animal Monitoring System (Columbus Instruments). Oxygen consumption (VO2), carbon dioxide production (VCO2) and ambulatory activity were measured over the course of 2 days. Respiratory exchange ratio (RER, or Respiratory quotient) and heat production were calculated but not reported because the assumptions are not met in abnormal metabolic states [35 (link)–37 (link)]. VO2 and VCO2 were normalized over grams of body weight. Food intake was calculated from the difference between the food allocated to every mouse at the start of the recording and the amount of food that was left at the end of the recording. All experiments were performed at the IBP Phenotyping Core Facility, University of Minnesota. Data were analyzed with unpaired t-test.

High-Fat Diet Induced Obesity in Mice

Check if the same lab product or an alternative is used in the 5 most similar protocols
Experiments were performed with six different batches of male C57Bl/6J mice (Charles River, Maastricht, the Netherlands). The batches differed in duration (4–34 weeks) and type of HFD (45% or 60% energy derived from lard fat; D12451 or D12492, Research Diet Services, Wijk bij Duurstede, the Netherlands) (electronic supplementary material [ESM] Table 1). Body weight was measured and lean and fat mass was assessed by MRI-based body composition analysis (Echo MRI, Echo Medical Systems, Houston, TX, USA). At the end of the diet intervention, mice were killed, perfused and organs were dissected for further analysis. All experiments were approved by the animal ethics committee of Leiden University Medical Center.

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
