Alcohol Dependence Induction Protocol

Alcohol dependence was induced by 9 days of daily intraperitoneal (i.p.) injection of EtOH (2 g/kg, 20% w/v in 0.9% saline, or saline control) with the alcohol dehydrogenase inhibitor fomepizole (4-methylpyrazole, 9 mg/kg, Sigma-Aldrich)(31 (link)). Blood alcohol level was assessed from tail bleeds 1h (intoxication) and 24h (withdrawal) after injection on day 6 or just before injection on day 7 using gas chromatography. One female from the naltrexone treatment group was euthanized following adverse effects from alcohol.
