1

Quantification of ATP and Cytotoxicity

Check if the same lab product or an alternative is used in the 5 most similar protocols
ATP in BALF was assessed using a Fluorometric Assay Kit (SIGMA ALDRICH, MO, USA). The LDH detection kit (Roche Diagnostics, Indianapolis, IN, USA) was used to assess the cytotoxic effect of different treatments. Data were expressed as the fold changes of different groups versus the control groups. Mouse CXCL1/KC ELISA kit (R&D Systems Inc. Minneapolis, MN, USA) was used to quantify KC in BALF.

GTP and ATP Quantification in Diabetic Mice

Check if the same lab product or an alternative is used in the 5 most similar protocols
BMCs and liver tissues were obtained from WT and db/db mice at 8 and 24 weeks. Equal amounts of protein were measured for GTP quantification using an enzymatic assay (ml027797, MLBio, Shanghai, China). ATP concentration was measured using a commercial Fluorometric Assay kit (213-579-1, Sigma-Aldrich, Saint Louis, US). All procedures were in accordance with the manufacturer’s instructions.

Fluorometric Assay for Glucose Uptake

Check if the same lab product or an alternative is used in the 5 most similar protocols
We used the glucose uptake Fluorometric Assay Kit from Sigma-Aldrich following the manufacturer’s instructions. Briefly, glucose uptake in C2C12 myotubes was measured using a glucose analog, 2-DG. 2-DG uptake was determined by a coupled enzymatic assay in which the 2-DG6P is oxidized, generating NADPH which reacts with the probe to generate a fluorometric signal (λex535/λem587 nm) proportional to the 2-DG taken up by the cells.

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
