Bdnf duoset elisa kit

Manufactured by R&D Systems
Sourced in Germany, United States

The BDNF DuoSet ELISA kit is a quantitative sandwich enzyme-linked immunosorbent assay (ELISA) designed for the measurement of human Brain-Derived Neurotrophic Factor (BDNF) concentrations in cell culture supernatants, serum, and plasma. The kit includes a capture antibody, a detection antibody, and a calibrator for BDNF.

Quantifying Serum BDNF Levels by ELISA

Check if the same lab product or an alternative is used in the 5 most similar protocols
Serum BDNF level was measured using BDNF DuoSet ELISA kit (DY248; R&D Systems, Wiesbaden-Nordenstadt, Germany) and DuoSet Ancillary Reagent kit 2 (DY008; R&D Systems) according to the manufacturer’s instructions. Briefly, sample or standard was added to a plate coated with capture antibody and the plate was incubated for 2 hours at room temperature. After washing the plate sufficiently, detection antibody was added to the plate and the plate was incubated for 2 hours at room temperature. Streptavidin conjugated to horseradish peroxidase was added to each well and the plate was incubated for 20 minutes at room temperature. Then substrate solution was added to each well for 20 minutes of incubation at room temperature following sufficient washing with wash buffer, and the plate was ready for determining the optical density at 450 nm wavelength using a microplate reader (SpectraMax iD3, Molecular Devices, Austria).

Biomarker Assessment in Biological Samples

Fasted blood was collected at T0 using red top BD Vacutainer® collection tubes (BD and Company, Frankin Lakes, NJ, USA). Tubes were left to clot at room temperature for ~50 minutes and subsequently centrifuged at 4000 r.p.m. for 10 minutes at 4ºC. The supernatant was aliquoted into vials and stored at -80ºC. Saliva samples were obtained using Salivettes (Sarstedt, Nümbrecht, Germany) and centrifuged at 4000 r.p.m. for 10 minutes and stored at -80ºC. The Hamilton Regional Medicine Program Core Laboratory quantified serum estradiol and progesterone levels using chemiluminescent microparticle immunoassay (Abbott Architect, Abbott Laboratories, Chicago, USA), serum testosterone using a chemiluminescent enzyme immunoassay (Immulite 2000; Siemens, Erlangen, Germany), and salivary cortisol using the Cortisol ELISA Kit (Enzo Life Sciences, New York, USA). Serum BDNF was analyzed using human BDNF DuoSet ELISA kit (R&D Systems, Minneapolis, USA).

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
