Taxifolin Attenuates DSS-Induced Colitis in Mice

Thirty male Institute of Cancer Research (ICR) mice (aged 7–8 weeks, approximately 30 ± 2 g BW) were obtained from Liaoning Changsheng Biotechnology Co., Ltd. (Benxi, China). Taxifolin was purchased from Yuanye Bio-Technology Co., Ltd. (S31601, Shanghai, China). DSS was obtained from Sangon Biotech Co., Ltd. (A600160-0250, Shanghai, China). The mice were randomly separated into three groups: Control, DSS, and Taxifolin + DSS (Taxifolin) group (n = 10). The mice from the Taxifolin group received Taxifolin dissolved in distilled water via intragastric gavage daily from day 1 to day 14 (100 mg/kg BW). On day 8, the mice from DSS and Taxifolin groups were given distilled water containing 3% DSS (w/v) for additional 7 days. The dose of Taxifolin is a safe dose to use in clinical studies (14 (link)). All the mice received a basal diet and were kept in cages in a room with a 12-h light and 12-h dark with controlled temperature (25 ± 2°C) and humidity (50 ± 5%). On day 15, all the mice were sacrificed. The animal experiment was performed under the guidelines of the Laboratory Animal Ethics Committee of Huazhong Agricultural University.

Immunomodulatory Effects of Taxifolin

The experimental protocols were approved by the Animal Ethics Committee of Laboratory Animals of the Institute of Laboratory Animal Resources of Tianjin (Tianjin, China).

(Shanghai, China). Thermo Fisher Scientific (Waltham, MA, USA) provided the methanol (HPLC quality), formic acid (99.9%, LC-MS grade), and acetonitrile (LC-MS grade). Cellulase (T richoderma Vride G, 400 u/mg), hemicellulase (Aspergillus niger, 20 u/mg), and pectinase (Aspergillus niger, 500 u/mg) were obtained from Yuanye Bio-Technology Co., Ltd. (Shanghai, China).

Analytical Characterization of Botanical Compounds

Quantification of Flavonoids in Acer nephrolepis

The reference substances taxifolin and quercetin, with purity above 98%, were obtained from Shanghai Yuanye Bio-Technology Co., Ltd. (Shanghai, China), and diosmin, with purity above 99%, was purchased from Ziyi Chemical Reagent Factory (Shanghai, China); chromatographic grade acetonitrile and phosphoric acid were obtained from Thermo Fisher Scientific (Shanghai, China). A 0.45 µm filtration was needed for the standards, flavonoid extract samples and eluent, and some samples were diluted to appropriate concentrations by pure water before HPLC analysis.
Leaves and bark of A. nephrolepis were provided by the Liangshui Experimental Forest Farm (Northeast Forestry University, Yichun, Heilongjiang, China) and collected from one tree of ten years of growth in September 2017. The leaves and bark of A. nephrolepis specimens were certified by Prof Huiyan Gu (Northeast Forestry University, Harbin, China). The materials were dried at ambient temperature for 2 weeks, ground into a homogeneous powder, and then sieved (60–80 mesh). The sieved samples were stored in polyethylene zipper bags at 4 °C until use. The same batch of sample was used in all experiments.

For chromatographic analysis, high-performance liquid chromatography (HPLC)-grade formic acid, acetic acid, and acetonitrile were obtained from Alfa Aesar (Shanghai, China) and Merck (Darmstadt, Germany). Ultrapure water was obtained from a Milli-Q system (Millipore, Bedford, MA, United States).

Ultrapure water supplied by a Milli-Q system (Millipore, Bedford, MA, USA) was used throughout the experiments.
