Actinobacteria Metabolites Production Protocol

Four actinobacterial strains were selected for extracts generation following phylogenetic analysis. A volume of 5 mL SV2 media (for 1 L, add 1 g calcium carbonate (Sigma-Aldrich, St. Louis, MO, USA), 15 g glucose (1st BASE, Singapore), 15 g glycerol (VWR, Radnor, PA, USA), and 15 g soya peptone (Oxoid, Hampshire, UK), pH adjusted to 7.0) was used to culture strains at 28 °C for 3 days under constant agitation at 200 rpm to generate a seed culture. The seed cultures were then inoculated in a 1:20 volume into five in-house liquid media (CA02LB, CA07LB, CA08LB, CA09LB, and CA10LB) as shown in Table 5. These media have been formulated and optimized by the Natural Product Library group at SIFBI for actinobacteria secondary metabolites production. The cultures were incubated for 9 days at 28 °C in the dark with shaking at 200 rpm. Following incubation, the cultures were lyophilized. The dried cultures underwent extraction using methanol (MeOH) and were subsequently filtered through Whatman Grade 4 filter paper. MeOH was then evaporated under reduced pressure to generate the crude extract.

Preparation of Polymeric Nanoparticles for Drug Delivery

Nanopure water (18 MΩ cm) was acquired by means of a Milli-Q water filtration system, Millipore (St. Charles). SGF (pH ~1.2) was made by dissolving 2 g NaCl and 8.3 ml concentrated HCL in nanopure water and adjusting to 1000 ml.

Haem-Mediated Colon Carcinogenesis Diet

All diets were based on a modified low calcium AIN76 diet in a powdered form (UPAE, INRA, France), balanced in iron (haem in haem-enriched diets vs. ferric citrate in control diets), proteins (20% using casein), and lipids (5% using safflower oil). Haem groups (HEM and HEM-Ca) received 1.5 μmol/g diet of haemin (Sigma Chemical), and control groups (CON and Ca) received 0.036% of ferric citrate to balance iron level. Calcium level is critical for the haem promotion of carcinogenesis; calcium was thus excluded from mineral mix but dibasic calcium phosphate was included in all diets at a low concentration of 3.4 g/kg. Calcium groups (Ca and HEM-Ca) received 2% of calcium carbonate (Sigma chemical) in order to chelate haem in the intestinal lumen.
