Quantifying Lung CXCL1 Levels in Mice

Following blood withdrawal, the animals were perfused with 20 ml PBS via the inferior caval vein. The lungs were then removed and snap-frozen in liquid nitrogen. Lung tissue was homogenized in a protein lysis buffer at 4°C, followed by centrifugation for 30 min at 4°C and 20,000 × g. Subsequently, supernatants were stored at −80°C. CXCL1 concentrations in the lungs were determined using a Mouse CXCL1/KC ELISA kit from R&D Systems according to the manufacturer's instructions (Wiesbaden-Nordenstadt). ELISA was performed using an Infinite M200 microplate reader (Tecan Deutschland GmbH).
