PLGA-based Controlled Drug Release

Poly [D,L-lactide-co-glycolide (PLGA)] 7525 7E (lactide/glycolide ratio, 75/25; molecular weight, 113,000 Da; inherent viscosity, 0.74 dl/g) was obtained from Lakeshore Biomaterials, Inc. (Birmingham, AL, USA). Butyl stearate was purchased from TCI Shanghai Development Co., Ltd. (Shanghai, China). Methylene dichloride was purchased from Beijing Chemical Works (Beijing China), and polyvinyl alcohol (PVA; Mw 13,000–23,000) was supplied by Sigma-Aldrich (Merck KGaA, Darmstadt, Germany). Isoperidone was synthesized in our laboratory. Sodium-dodecyl sulphate (SDS) was purchased from Baotaike Bio-Technology Co., Ltd, Shenyang, China.
Healthy female beagle dogs aged 1–2 years and weighing 12±2 kg were supplied by Yadong Laboratory Animal Research Center (Nanjing, China). The animals were kept in iron cages at 20±4°C, with 45–65% relative humidity and a 12 h light/dark cycle. They were fed with granulated feed and had free access to water. All animal experiments were approved by Institutional Animal Care and Use Committee of Jilin University (Changchun, China).

Formulation and Characterization of Exenatide-Loaded PLGA Microparticles

Exenatide was purchased from American Peptide Company, Inc. (Sunnyvale, CA, USA). PLGA 50/50 was obtained from Boehringer-Ingelheim (Resomer 503H, Ingelheim am Rhein, Germany). Sucrose, lysine, phenylalanine, proline, and poly(vinyl alcohol) (PVA, M.W. 13,000–23,000) were purchased from Sigma Aldrich (Saint Louis, MO, USA). Poloxamer 188 was kindly supplied by BASF (Lutrol®F68, Ludwigshafen, Germany) and 1,2-dimyristoyl-sn-glycero-3-phosphocholine (DMPC) was obtained from Corden Pharma (Liestal, Switzerland). Acetonitrile, trifluoroacetic acid (TFA), and dichloromethane (DCM) were purchased from Merck (Fair Lawn, NJ, USA). All other chemicals were of reagent grade and used without further purification.

Rosiglitazone was purchased from Cayman Chemical (Ann Arbor, MI). Dichloromethane (DCM) and polyvinyl alcohol (PVA, Mw 13,000–23,000, 87–89% hydrolyzed) were purchased from Sigma (St. Louis, MO). Dulbecco’s Modified Eagle’s Medium (DMEM) with 4.5 g/L glucose and L-glutamine was purchased from Corning Cellgro (Corning, NY). Trypsin with 0.25% EDTA, Fetal Bovine Serum (FBS) and Pen/Strep were purchased from Fisher (Hampton, NH). Super Calf Serum was purchased from GemCell (Sacramento, CA). Insulin (bovine), dexamethasone, and methylisobutylxanthine (IBMX) were purchased from Sigma (St. Louis, MO). 10% Neutral Buffered Formalin was purchased from Sigma (St. Louis, MO). Nile Red was purchased from Molecular Probes (Eugene, OR). Adiponectin ELISA kit was purchased from R&D systems (Minneapolis, MN).

Preparation of PLGA-based Nanoparticles

High-purity LFD (Figure 1) was kindly donated by Pharmathen S.A. (Athens, Greece). CS of low molecular weight (50–190 kDa, degree of deacetylation 75–85%) and sodium tripolyphosphate (TPP), used as ionic crosslinker, were purchased from Sigma-Aldrich Co. (Steinheim, Germany). [2-(methacryloyloxy)ethyl]dimethyl-(3-sulfopropyl)ammonium hydroxide (SDAEM) with 95% purity was purchased from Fluorochem (London, UK), while potassium persulfate (K2S2O8, KPS, purity 99.0%) was purchased from Merck (Athens, Greece). L-lactide (98+%) (S,S)-3,6-Dimethyl-1,4-dioxane-2,5-dione was purchased from Alfa Aesar Chemicals (Kandel, Germany). Methoxy poly(ethylene glycol) (average Mw 2000), tin(II) 2-ethylhexanoate (TEH) (96%) catalysts, and poly(vinyl alcohol) (PVA, Mw 13,000–23,000, 87–89% hydrolyzed) were all obtained from Sigma-Aldrich chemical company (Saint Louis, MO, USA). PLGA 75/25 (PURASORB® PDLG 7507) was purchased from Corbion (Gorinchem, The Netherlands). All other solvents or reagents used were of analytical grade and purchased also from Sigma-Aldrich.

Acetone, dichloromethane (DCM), MTT and Triton X100 were bought at Fisher Scientifics (USA), Macron Fine Chemicals (USA), Roche (USA) and Research Product International (USA), respectively. Macrophages Raw 264.7 (ATCC® TIB-71™) was obtained from ATCC (USA).
