Strains of Escherichia coli ATCC 25922, Staphylococcus aureus ATCC 29213, and Candida albicans ATCC 14053 were obtained from American Type Culture Collection (ATCC). Glutaraldehyde, paraformaldehyde, sodium cacodylate, PBS, and solvents used in this study were from Sigma-Aldrich (San Luis/MO, USA). Nutrient agar, Müeller-Hinton broth, Sabouraud dextrose agar, and Sabouraud dextrose broth were from Himedia (Mumbai, India).

Oral Candida Strains Collection

Antifungal Activity of Peptaibol Extract

The antifungal activity of a crude peptaibol extract was measured by the disc-diffusion method. Discs with a 6 mm diameter containing 40 µL of the sample were deposited on PDA agar plates (Sigma-Aldrich, St. Louis, MO, USA). The diameter of the inhibition zones was measured after 24 h at 28 °C. Amphotericin B solution (Sigma-Aldrich) was used as a positive control. The minimal inhibitory concentration (MIC) value of each individual compound was determined using the broth two-fold microdilution method according to CLSI/NCCLS documents M27-A3, M38-A, and M38-A2 [54 ]. Yeast strains Candida albicans ATCC 14053 and the fungal strain Aspergillus niger ATCC 16404 were obtained from the American Type Culture Collection (ATCC, Manassas, VA, USA).

Antifungal Activity of Peptides

Fungal strains were purchased from the American Type Culture Collection (ATCC, Manassas, VA, United States) as follows: Aspergillus brasiliensis ATCC 9341 and Candida albicans ATCC 14053 strains. Briefly, the cell suspension of both fungal species was adjusted to 1.0 × 105 CFU/mL in buffered peptone water (BPW) (bioMerieux, Florence, Italy). Peptide stock solutions in DMSO were added to the cell suspension at a final concentration of 25 μM and 50 μM and incubated for 6 h a 37°C. The minimum fungicidal concentration (MFC) was determined by plating 100 μL cultures on DG18 plates (Dichloran 18% Glycerol Agar—ISO 21527-2) for CFU counting. After 7 days at 25°C, the MFC was defined as the lowest peptide concentration that resulted in 99.9% killing compared with the drug-free group. The analyses were performed in triplicate on three different experiments.
