Extraction and Analysis of Saussurea laniceps

Saussurea laniceps (SL) was collected in Tibet (China) in September, 2014. The material was identified according to macroscopic and microscopic features40 (link). Voucher specimens (No. MT-11) were deposited in the Chinese Medicines Center, Hong Kong Baptist University.
Ethanol was purchased from Merck (Darmstadt, Germany). Acetonitrile (ACN) and formic acid of HPLC grade, n-butanol, ethyl acetate, mEthanol, chloroform of analytical grade were purchased from Lab-scan (Bangkok, Thailand). Hydroquinone with a purity of 99.5% was purchased from Phytomarker Ltd. (Tianjin, China). Methyl tert-butyl ether (MTBE) and other chemicals of analytical grade were purchased from Sigma (St. Louis, MO). The water used in the experiments was collected from a Mili-Q ultrapure water system (Bedford, MA, USA).

Antioxidant Capacity Evaluation of Catechin

All chemicals and solvents were analytical grade. Standard catechin was purchased from Sigma® (Sigma‐Aldrich, USA). Stock solutions of Standard catechin were freshly prepared for daily use. Stationary phase was TLC plates precoated with silica gel 60 F254 (20x10 cm) and thickness of 0.2 mm purchased from Merck (USA). Ethanol, hexane, dichloromethane, ethyl acetate, and mEthanol were purchased from Labscan (Dublin, Ireland). 2,20‐azino‐bis‐3‐ethylbenzothiazoline‐6‐sulfonic acid (ABTS), 2,4,6 tripyridyl‐s‐triazine (TPTZ), 6‐hydroxy‐2,5,7,8‐tetramethylchroman‐2‐carboxylic acid (Trolox), and linoleic acid were purchased from Sigma‐Aldrich (St. Louis, MO, USA). Tris base was purchased from Fisher Chem Alert (Fair Lawn, NJ, USA). Potassium persulfate, ferric chloride, ferrous sulfate, and sodium acetate were purchased from Fisher Chemicals (Loughborough, UK). Quantitative Analysis of Metabolites in Fermented Media

Aliquots collected during the incubation of isolated strains, were extracted and analyzed by HPLC-DAD-ESI-Q (MS) as previously described (García-Villalba et al., 2016 (link)). Briefly, fermented medium (5 ml) was extracted with ethyl acetate (5 ml) (Labscan, Dublin, Ireland) acidified with 1.5% formic acid (Panreac), vortexed for 2 min and centrifuged at 3500 g for 10 min. The organic phase was separated and evaporated and the dry samples were then re-dissolved in methanol (250 μl) (Romil, Barcelona, Spain). An HPLC system (1200 Series, Agilent Technologies, Madrid, Spain) equipped with a photodiode-array detector (DAD) and a single quadrupole mass spectrometer detector in series (6120 Quadrupole, Agilent Technologies, Madrid, Spain) was used as previously described (García-Villalba et al., 2016 (link)). Calibration curves were obtained for EA, urolithin C as well as urolithin M6 and isourolithin A (Villapharma SL, Murcia, Spain) with good linearity (R2 > 0.998). Isourolithin A and urolithin C were quantified at 305 nm, while urolithin M5, urolithin M6, and EA were quantified at 360 nm, all with their own standards.

Sildenafil Citrate and Reference Analysis

Sildenafil citrate and reference standard (potency 99.4% as is) were obtained from Smilax Laboratories Limited (Hyderabad, India). Aspirin and BZA were purchased from P. C. Drug Center Co. Methanol, ethyl acetate, and all reagents were supplied by Labscan (Bangkok, Thailand). Potassium bromide and other excipients were purchased from PC Drug Center Co., Ltd. (Bangkok, Thailand). All chemical compounds and all reagents were used as received except sildenafil (base) was prepared in the laboratory.

Evaluating Esterase Activity in LAB

The activity of the L. plantarum esterase genes expressed in several lactic acid bacteria was assayed by incubating the bacteria in the presence of hydrozybenzoic (methyl gallate and gallic acid) and hydroxycinnamic (methyl ferulate and ferulic acid) derivative compounds (Curiel et al. 2010) . Bacterial GM17 or MRS culture media containing a phenolic compound (1 mM final concentration) were inoculated with the corresponding bacterial culture and incubated for seven days at 30 C in aerobic or anaerobic conditions in presence of chloramphenicol or erythromycin (5 mg/ml), depending of the plasmid carried. After incubation, the phenolic compounds present in the supernatants were extracted twice with one third of the reaction volume of ethyl acetate (Lab-Scan, Ireland) and analysed by HPLC-DAD (Bartolom e et al. 2000) .

GC-ECD Analysis of Organochlorine Pesticides

Petroleum ether, (40-60°C) Analar (BDH, England), dichloromethane, (Jansen Chimica, Belgium), n-hexane (Riedel de-Hean, Germany), acetone, Analar (BDH, England), ethanol, (GCC, U.K.), diethyl ether(GCC, U.K.), ammonia solution (25%) ( Riedel de-Hean, Germany), cyclohexane (Scharlau, Spain), ethyl acetate (Lab-scan, Ireland), toluene ( Riedel de-Hean, Germany). Organochlorine pesticides (OCP's) used internal standards o,p-DDT, p,p-DDT, o,p-DDE, p,p-DDE, o,p-DDD, p, DDD, α-HCH, β-HCH, γ-HCH, aldrin, dieldrin, endrin, heptachlor, heptachlorepoxide, HCB, and isodrin have been obtained from Dr. Ehrebstorfer GmbH laboratory (Germany). Analyticalgrade anhydrous sodium sulfate (Fluka, Switzerland) has been heated at 550°C for 3hrs and kept in a closed container.
