Adenosine, NECA, IB-MECA, HEMADO, DPCPX (8-cyclopentyl-1,3-dipropyl-7H-purine-2,6-dione) and MRS 1220 were purchased from Sigma-Aldrich and dissolved in dimethyl-sulphoxide (DMSO). CA200645, a high affinity AR xanthine amine congener (XAC) derivative containing a polyamide linker connected to the BY630 fluorophore, was purchased from HelloBio (Bristol, UK) and dissolved in DMSO. AV039, a highly potent and selective fluorescent antagonist of the human A3R based on the 1,2,4-Triazolo[4,3-a]quinoxalin-1-one linked to BY63039 (link), was kindly gifted to us by Stephen Hill and Stephen Briddon (University of Nottingham). The concentration of DMSO was maintained to < 1.5% across all experiments (1.26% for all cAMP assays, 1% for pERK1/2 assays and 1.02% or 1.1% for NanoBRET ligand-binding experiments using CA200645 or AV039, respectively).

Combination Therapy for Cancer Treatment

Mice were given 400 mg/kg/dose (100µl total volume) of D/L-1MT (Sigma) diluted in Methocel/Tween (0.5% methylcellulose (w/v), 0.5% Tween 80 (v/v) in water) twice daily by oral gavage (p.o.); (33 (link)) 1, 10, or 25 mg/kg/dose (100µl total volume) of MTX (Hannah Pharmaceuticals) diluted in Methocel/Tween weekly p.o.; 0.5mg/kg IB-MECA (Sigma) diluted in saline daily i.p.; 10mg/kg theophylline (Sigma) diluted in Methocel/Tween daily p.o.; 1% difluoromethylornithine (DFM0; ILEX oncology) in the drinking water; 1 or 25mg/kg folinic acid (Sigma) diluted in Methocel/Tween daily p.o.; or a combination of 1MT, MTX, and the inhibitors. folinic acid and MTX were administered 8hr apart to avoid interference with their uptake (28 (link), 30 (link)). Control mice were given an equal volume of carrier alone (Methocel/Tween).
