Curcumin and THC Cytotoxicity Evaluation

Phosphate buffer solution (PBS), 0.1% tween-20 in phosphate buffer solution (PBST), and sodium dodecyl sulfonate (SDS) were purchased from Zhengjiang Wanbang Pharmaceutical Co. (Wenling, China). The primary antibodies against Bax, Bcl-2, poly (ADP-ribose) polymerase (PARP), cytochrome c, and the horseradish peroxidase (HPR)-conjugated goat anti-mouse secondary antibody were provided by BioVision, Inc., (BioVisio, Palo Alto, CA, USA). All the other cell culture reagents were purchased from Sinopharm (Beijing, China), and all the other chemicals were of analytical grade.

Lonicera japonica Bioactivity Evaluation

All laboratory animal procedures were conducted in strict accordance with the National Institutes of Health Guidelines for the Use of Laboratory Animals. All procedures involving animals and their care have been approved by the Animal Ethics Committee of the Yijishan Hospital of Wannan Medical College (SCXK 2019-0007). All the mice were male between 8 and 12 weeks of age. The male mice were placed under standard conditions, with a 12-hour light/dark cycle and plenty of water and food. All efforts were made to minimize suffering. Male Balb/c mice were purchased from the Shanghai Laboratory Animal Center (Shanghai, China). Lonicera japonica was purchased from Zhongshan chemist's shop in Wuhu, China. Standard monosaccharides, including D-mannose, D-rhamnose, D-glucose, D-fucose, D-xylose, D-galactose, and D-arabinose were bought from Sigma (St. Louis, MO, USA). Dulbecco's modified eagle medium (DMEM), fetal bovine serum (FBS), dimethyl sulfoxide (DMSO), and 3-(4, 5-dimethylthiazol-2-yl)-2, 5-diphenyltetrazolium bromide (MTT) were purchased from Sigma–Aldrich (St. Louis, MO, USA). Annexin V-FITC was purchased from BioLegend, Inc. (San Diego, CA). AST, CPK, LDH, Caspase-3, Caspase-8, and Caspase-9 kits were purchased from Nanjing Jiancheng Bioengineering Institute (Nanjing, China). SOD, CAT, GSH-Px, and MDA kits were purchased from BOMEI Biotechnology Co., Ltd. (Hefei, China).

Butin-Mediated Cytoprotection Against Oxidative Stress

Butin was purchased from National Institute for the Control of Pharmaceutical and Biological Products, and the purity being more than 98%. Dulbecco’s Modified Eagle’s Medium (DMEM) and other cell culture supplies were purchased from Gibco (Life Technologies, Grand Island, NY). 3-[4, 5-Dimethylthiazol-2-yl]-2, 5-diphenyltetrazolium bromide (MTT) and 2, 3, 5-triphenyltetrazolium chloride (TTC) were obtained from Sigma (St. Louis, MO). The detection kits of malondialdehyde (MDA), superoxide dismutase (SOD), glutathione (GSH) and glutathione disulfide (GSSG) and caspase 3 were purchased from Nanjing Jiancheng Bioengineering Institute (Nanjing, P.R. China). Monoclonal rabbit anti-mouse P-AMPK, AMPK, P-GSK-3β, GSK-3β, Bcl-2, β-actin, Bax, NQO1, HO-1 and Nrf2 antibodies were purchased from Cell Signaling Technologies, MA, USA. All other chemicals used in this experiment were the purest grade commercially available.

Biochemical Markers of Knee Arthritis

Commercial ELISA kits were used to determine the levels of MDA (cat. no. BB-4709-1; BestBio), SOD (cat. no. BB-4710-2; BestBio), TNF-α (cat. no. H052; Nanjing Jiancheng Biological Engineering Institute, Nanjing, China), IL-6 (cat. no. H007; Nanjing Jiancheng Biological Engineering Institute) and caspase-3 (cat. no. BB-4106-2; BestBio) in knee joint arthritis samples, according to the manufacturer's protocols.
Statistical analyses. Data are presented as the mean ± standard deviation. SPSS 11.0 software (SPSS, Inc., Chicago, IL, USA) was used to perform one-way analysis of variance and Fisher's protected least significant difference test. P<0.05 was considered to indicate a statistically significant result.

Polyethylene Microplastic Effects on Marine Organisms

Polyethylene (PE) is one of the most commonly used plastics globally, and commonly seen in packaging. PE was selected because it is a common plastic polymer used in microplastics sampling in marine and beaches (Ding et al., 2019; Jensen et al., 2019) . Polyethylene microplastic was purchased from Yineng Platic Materials CO., Ltd. (Dongguan, China). Polyethylene suspension was prepared by ultraviolet disinfection of aerated seawater. The composition of polyethylene was con rmed by Raman spectroscopy (RS, SR-510 Pro, Ocean optics Asia, 785 nm laser, Raman shift 50-3500 cm -1 ), and it showed in Fig S1 . Analytically pure acetone (AR 99.9% purity), chromium chloride hydrate of analytical grade (2:5) (99.0% purity) were purchased from Macklin Chemical Co., Ltd. (Shanghai, China). In ltered sea water, 100 μg/L and 50 μg/L Cr(III) stock solutions were prepared. Assay kits for measurement of levels of NDA + /NADH, G6DPH, LDH, Caspase-3 and Caspase-1 were purchased from Nanjing Jiancheng Bioengineering Institute (Nanjing, China). BCA protein assay kit was offered by Beyotime Institute of Biotechnology (Jiangsu, China). The seawater used in the whole experiments was arti cial.

Apoptosis Markers in Rat Synovium

The rats were sacrificed and then their synovial tissues were isolated from their knee joints and embedded in paraffin. Sections (5-µm thick) were cut on a microtome and then dewaxed in water. Endogenous peroxidases were inactivated and antigen retrieval was conducted using a boiling method. Then, the sections were blocked in goat serum for 15 min at room temperature and incubated overnight at 4°C with antibodies against Bax (1:100) and Bcl-2 (1:100) (both from BBI Life Science Corporation), and caspase-3 (1:100; Nanjing Jiancheng Bioengineering Institute). A biotin-labeled secondary antibody and SABC solution were added dropwise, successively and incubated for 20 min at room temperature. The sections were developed with DAB, re-stained with hematoxylin, dehydrated, brightened and mounted with neutral gum. Images were captured on a light microscope (Olympus) and OD values were calculated with IPP software.

The antibodies, including AMPK, p-AMPK, β-actin, and GLUT4, were purchased from Cell Signaling Technologies, Inc. (Danvers, MA, USA). The secondary antibody (anti-rabbit IgG-B; cat. no. sc-53804) was purchased from Santa Cruz Biotechnology, Inc. (Dallas, TX, USA).
