For HPLC and LC-MS/MS analysis, acetonitrile, methanol, and formic acid (HPLC grade) were obtained from Avantor Chemicals (Radnor, PA, USA), trichloracetic acid and heptafluorobutyric acid were obtained from Sigma Aldrich (St. Louis, MO, USA).

Permeability Assay for Compound Transport

According to the established method of permeability assay35 (link), 36 (link), on Day 28, 100 μl of the test compounds (1 μM propranolol, 10 μM mannitol or 10.3 μM caffeine) were added to the apical side of the transwell membrane with a confluent monolayer of hIECs or Caco-2 cells and the transport of the compound across the monolayer was monitored over an 8 hour time period by taking the medium samples from the apical and basolateral compartments at 1 hour, 2 hours, 4 hours and 8 hours. The compound concentrations in the sample medium were measured using Propranolol ELISA test kit (Neogen), D-mannitol Colorimetric assay kit (Sigma) and Caffeine/Pentoxifylline ELISA test kit (Neogen), respectively.
