Cytokine-Restimulated T Cell Analysis

For cytokine restimulated human and mouse T cells, in some instances cells grown in normal media or tumor supernatant were treated with cycloheximide for 30 minutes prior to being restimulated for 4 hours with Cell Stimulation Cocktail and GolgiPlug Brefeldin A (eBioscience). For tumor infiltrating T cell analysis, single cell suspensions were counted and plated at 5x105 cells per well and stimulated with Cell Stimulation Cocktail and GolgiPlug Brefeldin A for 3 hours at 37C. Samples were then stained with Zombie NIR viability dye (Biolegend) followed CD45.2-BV510 (Biolegend, RRID:AB_2561393) and CD8-Alexa Fluor 647. IFNγ-PE (Invitrogen, RRID:AB_466192) and TNF-α-FITC (Invitrogen, RRID:AB_465418) intracellular FACS staining was performed using the Foxp3/Transcription Factor Fixation/Permeabilization Concentrate and Diluent kit and Permeabilization Buffer (eBioscience). Samples were run on a BD Accuri C6 flow cytometer or a Cytek Northern Lights and analysis was performed with FlowJo software.

Lung CD8+ T Cell Phenotyping

Lung CD8+ T cells were washed in PBS containing 0.5% bovine serum albumin and 2 mM EDTA, then incubated with the following antibodies: CD127 (A7R34) PE (eBioscience 12-1271-81), Klrg1 (2F1) APC (eBioscience 17-5893-81), CD4 (RM4-5) PerCP-Cy5.5 (eBioscience 45-0042-82), Tcrβ BV605 (BioLegend 109241), CD45.2 BV510 (BioLegend 109837), and Cd8a BV711 (BioLegend 100747). Detailed Immunological Assays for LB27.4 Cell Line

The I-Ab+ B cell lymphoma line LB27.4 was purchased from the American Type Culture Collection. The following FACS antibodies were used in this study: Fc Block, CD8a BV 421, CD45.2 BV 510, B220 BV 605, CD127 AF 488, CD45.1 PE, KLRG1 PE-Cy7, CD25 AF 647, TNF-α AF 488, and IFN-γ AF 647 (all from Biolegend). Anti-alpha-adducin (ab40760) from Abcam was used for western blots. Anti-PKC-θ (C-18), anti-alpha-adducin (H-100; used for microscopy), and anti-phospho-alpha-adducin (Ser 726) antibodies were from Santa Cruz Biotechnology. CellTrace Violet, CellTrace CFSE, and AF 568-labeled phalloidin were from ThermoFisher. Class I (257-264) and Class II (323-339) ovalbumin peptides were from AnaSpec. GolgiPlug was from BD Biosciences. Anti-CD3 (145-2C11) and anti-CD28 (37.51) were from Bio-X-Cell.
