Biophysical Analysis of Cytochrome c Interactions

Cyt c (from equine heart), phosphatidylcholine (eggPC, from egg yolk), 3-Mercaptopropionic acid, 6-Mercaptohexanol, potassium chloride (KCl), sodium l-ascorbate, ethanol, 1-Octadecanethiol (ODT), trifluoroacetic acid (TFA), acetonitrile (ACN), and sinapinic acid were purchased from Sigma (St. Louis, MO). All aqueous solutions were prepared using ultrapure water (18.2 MΩ cm) from a Milli-Q purification system (Millipore, Burlington, MA). A CL binding dye, 10-nonyl acridine orange (NAO), was obtained from Enzo Life Sciences (Farmingdale, NY).

Micro-Scale Friction Characterization of Mixed SAMs

Clean gold surfaces were immersed in a 1 mM mixed thiol solution in ethanol containing 90% OEG-thiolate and 10% hexaethylene spaced carboxylic acid terminated alkanethiol (Sigma). After exposure the mixed SAM samples to different doses of laser through the interferometer, samples were immersed in a 2 mM solution of 1-octadecanethiol (Sigma) for 2 hours, rinsed in ethanol and dried with nitrogen. The samples were imaged by FFM. Friction force microscopy images were acquired in air using a Bruker MultiMode 8 NanoScope V AFM. The probes used for FFM were silicon nitride probes (NP series, Bruker) with spring constant k = 0.12 N m–1.

Functionalized Au Nanoparticles for RNA Extraction

Gold(III) chloride trihydrate (HAuCl4·3H2O, ≥99.9%), sodium citrate tribasic dihydrate (Na3Ct.2H2O, ≥99%), 1-octadecanethiol (98%, C18SH), poly(allylamine hydrochloride), (PAH, M.W. 15,000 g/mole), 4-(2-hydroxyethyl)-1-piperazineethanesulfonic acid (HEPES) were obtained from Sigma Aldrich and were used as received. 1-palmitoyl-2-oleoyl-sn-glycero-3-phospho-L-serine (sodium salt) (POPS), 1-palmitoyl-2-hydroxy-sn-glycero-3-phosphocholine (LPC) were obtained from Avanti Polar Lipids and were used as received. Trizol (Invitrogen) and RNeasy kit (Qiagen) were used in the extraction of RNA. Ultrapure deionized water (17.9 MΩ, Barnstead NANOpure II) was used for all solution preparations. Glassware was cleaned with aqua regia and rinsed thoroughly before use. Absorption spectra of Au NPs (Figure S1) were taken on a Cary 500 scan UV-vis-NIR spectrophotometer and absorption spectra of RNA were taken on Nanodrop 1000. Zeta potential and dynamic light scattering measurements were performed on a Brookhaven Zeta PALS instrument.

Synthesis and Characterization of Organometallic Compounds

Silicon wafers with a 5 nm Ti adhesion layer and a 100 nm Au layer were obtained from Platypus Technologies (New Orleans, LA). Ge (100) wafers were obtained from MTI Corporation (Richmond, CA).

Doxorubicin Derivatives Nanoparticle Synthesis

Reconstitution of Cholera Toxin Receptors

BK-7 glass substrates were from Corning (Painted Post, NY). Chromium and gold used for electron-beam evaporation were acquired as pellets of 99.99% purity from Kurt J. Lesker (Jefferson Hills, PA).

Liposome-Coated Gold Nanoparticle Synthesis

A total amount of 2 μmole of lipid mixture (Avanti Polar Lipids) was prepared by mixing DPPC (10 mM), cholesterol (10 mM), DSPE-PEG (2000) azide (3 mM), and DOPS (5 mM) in a clean 25 mL round-bottom flask. The solvent chloroform in the lipid mixture was removed by rotary evaporator. A uniformly thin lipid layer formed on the bottom of the flask, which was then further dried under vacuum overnight. HEPES buffer (20 mM, Sigma-Aldrich, wt 238.30 g) was added to the dried lipid layer (1 mL 10 mM HEPES buffer per 1 μmole lipid), and the mixture was sonicated for 5 min to form a clear liposome solution. For each 500 μL of liposome solution, 50 μL of Au NP pellet (2×1011 particles/mL) was added. Subsequently, 40 μL of 1-Octadecanethiol (8.72 mM, Sigma-Aldrich, wt 286.56 g) dissolved in ethanol was added. The final volume was brought to 1 mL with HEPES buffer. The mixed solution was then shaken for 5 h, 18 h or 24 h, respectively. The NP were subsequently washed three times through centrifugation (720g, 10 min) and re-suspended in HEPES buffer.

Fabrication of Thiolated Self-Assembled Monolayers
