Handelin Extraction and Characterization

Handelin (C32H40O8; molecular weight 552.6552) was obtained from Baoji Herbest Bio-Tech Co., Ltd. (Shanxi, China) and affirmed by H1NMR and MS data. The purity was determined to be > 98% by HPLC method. Fatty acid-free bovine serum albumin (BSA) was from Equitech-Bio, Inc. (Kerrville, TX, USA). Palmitic acid (PA, TCI, Shanghai, China) was dissolved in absolute ethanol as stock solution and then conjugated with 10% BSA to achieve a concentration of 100 μM as previously described (Li et al., 2015 (link)). Investigating BAMBI and CTGF Knockdown in Multiple Myeloma

ON-TARGETplus Human CTGF siRNA-SMART pool, ON-TARGETplus Human BAMBI siRNA-SMART pool, and ON-TARGETplus non-targeting Pool siRNA were purchased from Dharmacon (Lafayette, CO, USA) and were used for all transfection experiments. BAMBI siRNA(h) #sc-60243, a pool of 3 target-specific siRNAs (Santa Cruz Biotechnology, Dallas, TX, USA) was only used to confirm the effect of knockdown BAMBI expression on MM cell proliferation. An antibody against BAMBI/NMA (ab203070) was purchased from Abcam (Cambridge, UK), anti-CTGF (sc-14939) and anti-goat IgG (sc-2354) from Santa Cruz Biotechnology (Dallas, TX, USA), and antibodies against cyclin D1 (92G2), cyclin D3 (2936), CDK2 (2546), CDK4 (1279), phospho-Smad2 (Ser465/467), phospho-Smad3 (Ser423/425) (C25A9), Smad2/3, rabbit IgG (7074), and mouse IgG (7076) from Cell Signaling Technology (Danvers, MA, USA). Anti-β-actin (A5441) was obtained from Sigma-Aldrich (St. Louis, MO, USA).

Protein Expression Analysis Protocol

Tissues or tumor cells were lysed with 2% sodium dodecyl sulfate (SDS) and the protein concentration was then measured with BCA protein assay kit. Next, proteins were separated by SDS-PAGE gel and transferred to a nitrocellulose membrane. After blocking, the membrane was incubated with different primary antibodies and secondary antibody (with washing between steps). Finally, the target protein bands were visualized via chemiluminescence system.
