MTLn3 cells were seeded onto acid washed 25 mm glass cover slips (Fisher) coated with 40 µg/ml Rat Tail Collagen 1 for 1 hour at room temperature. Cells were allowed to attach overnight and then were subsequently starved for 3 hours in L15 medium with .35% BSA (Life Technologies). The glass cover slip was then transferred to Attofluor live cell imaging chambers (Life Technologies), mounted onto the microscope, and stimulated with 100 ng/ml EGF or 1 µg/ml preclustered ephrin b1-fc. Ephrin b1-fc was preclustered with a 1/10 concentration of Goat anti-human fc for 1 hour at 37 °C. In some experiments, EGF treatment was followed by the addition of 1 µg/ml preclustered ephrin b1-fc or 1 µg/ml preclustered fc.
