Naïve CD4+ T Cell Transfer Colitis

Naïve CD4+ T cell were isolated from C57BL/6 (CD45.1) mouse spleens and lymph nodes with a mouse CD4+ T cell isolation kit as per the manufacturer's instructions (Stemcell Technology, Canada). The isolated CD4+ T population was labeled with CD45Rb-FITC, CD62L-PE, and CD4-APC (eBiosciences, San Diego, CA). CD4+CD62L+CD45RBhi T cells were sorted by flow cytometry. Then 3.5 × 105 cells were injected into RAG1−/− mice by intraperitoneal (IP) injection. Mouse body condition, the form of their stool and weight were monitored at least weekly. Mice started to lose weight and develop looser stools around 3–4 weeks in our facility indicating that colitis was present.

Multiparameter Flow Cytometry of Murine Immune Cells

The anti-mouse antibodies B220-PE-Texas Red, CD4-APC-CyC7, CD8-PerCP-Cy7, Foxp3-APC, CD11b–Pacific Blue, CD45RB –FITC, CD138-APC, IgA-FITC, anti-α4β7-FITC and CCR9-PerCP-Cy7 and their respective fluorochrome-conjugated isotypes were purchased from eBioscience (San Diego, CA). Intracellular Foxp3 staining was performed using the Foxp3-Transcription Factor Staining Buffer Set (eBioscience, San Diego, CA) as per manufacturer’s recommended protocol. Cells were acquired on an LSRII flow cytometer (BD Biosciences, San Diego, CA) or a CyAn™ ADP Analyzer (Beckman, Coulter, Inc., Carlsbad, CA) and data were analyzed using FlowJo software (Tree Star, Ashland, Or).
