Formulation and Characterization of PEG-PLGA Nanoparticles

Poly (ethylene glycol)-methyl ether-block -poly (lactide-co-glycolide) (PEG-PLGA50:50, MnPEG 2000 Da and 10,000 Da MnPLGA), dimethyl sulfoxide (DMSO), and coumarin 6 dye were purchased from Sigma Aldrich (St. Louis, MO, USA). Rhodamine 6G dye, dichloromethane (DCM), polyvinyl alcohol (PVA) (87–89% hydrolyzed, high molecular weight), acetonitrile, phosphate buffer saline (PBS), polyvinyl chloride (PVC) tubing, syringes (glass and plastic), syringe needles, and polytetrafluoroethylenes (PTFE) based consumables such as syringe filters (0.45 μm), evaporating dish, and magnetic stirring bars were obtained from Fisher Scientific. All other chemicals and reagents used in this study were of analytical grade.

Resveratrol and Thymol Formulation Evaluation

Resveratrol (RSV) (high purity > 98%) was purchased from Guangzhou Phytochem Sciences Inc. (Guangzhou, China). Thymol was purchased from Oxford Lab Fine Chem I.I.P (India). Dialysis bags (molecular weight cut off 12,000-14,000 Da) (Serva, Heidelberg, Germany). Coumarin-6 dye was obtained from Sigma-Aldrich (St. Louis, MO, USA). 3-(4,5-dimethylthiazol-2-yl)-2,5diphenyltetrazolium bromide (MTT) was supplied by SERVA Electrophoresis GmbH (Germany). TRIzol ® Reagent was provided by Invitrogen, Thermo Fisher Scientific (Waltham, MA, USA). The primers were acquired from Sigma-Aldrich (UK) and a one-step RT qPCR kit (SYBR Green with low ROX) was purchased from Intron Biotechnology, Korea. B-cell Lymphoma/Leukemia 2 ELISA kit was purchased from Novus Biologicals, UK (NBP2-69946), and the Nuclear Factor Kappa B ELISA kit was obtained from Wuhan Fine Biotech Co., Ltd (Wuhan, Hubei, China). Acetonitrile and DMSO (HPLC grade) were purchased from Fischer Scientific (Loughborough, UK). Methanol, chloroform, ethanol, and formalin were provided by Al-Gomhureya Co. (Cairo, Egypt). All other reagents and chemicals were of analytical grade.

Visualizing Nanoparticle-Cancer Cell Interactions

Nanoparticle interactions with cancer cells were observed using confocal microscopy. HCT116 colon cancer cells and PC3 prostate cancer cells were seeded at a density of 3 × 105 cells/well in a four-well tissue culture slide (BD Biosciences, Bedford, MA). The cells were incubated for 24 hours at 37°C. Cells were treated with 625 ng of PLGA nanoparticles containing Coumarin 6 dye (Sigma Aldrich, St. Louis, MO) and were incubated for an additional 2, 4, or 24 hours. Cells were stained with DRAQ5 (Life Technologies, Grand Island, NY) nuclear staining dye after incubation with the particles. Nanoparticle interactions were observed using a Zeiss Confocal Microscope (Zeiss, Peabody, MA).
