Cell Fixation and Flow Cytometry

Trypanosome Cell Cycle Analysis

Trypanosomes (1x105/ml) were cultured for 24 hours with deferoxamine at 2X IC50, gallic acid at ½ IC50, diminazene aceturate at ½ IC50 and harvested by centrifugation at 2700 rpm, then washed twice with PBS and fixed in 70% cold ethanol at -20°C overnight. The trypanosomes were then washed twice in PBS and 200 μl of guava cell cycle reagent (Sigma-Aldrich) was added and incubated in the dark for 30 minutes. Cell cycle analysis was done using the LSRFortessa X-20 flow (BD Biosciences) and FlowJo v10 software. At least 5000 trypanosomes were counted for each measurement.
