MTT-based Cell Viability Assay

Check if the same lab product or an alternative is used in the 5 most similar protocols
The viability of PTX-resistant SKOV3 and HeyA8 was analyzed using MTT detection kit (Beyotime, Jiangsu, China). In brief, cells were seeded in 96-well plates (5 × 103 per well) and cultured for 16 h. The cells were treated with test compounds for the defined time, and MTT solution (Beyotime) was added into each culture well. Four hours later, dimethyl sulfoxide (Millipore) was adopted to dissolve formazan. Finally, cell viability was analyzed using microplate reader (Thermo Labsystems, Waltham, MA, USA) with absorbance at 570 nm.

MTT Assay for Cell Viability

Check if the same lab product or an alternative is used in the 5 most similar protocols
MTT detection kit (C0009, Beyotime, China) was used in this section. MTT solution (10 μL) was added to each well and the cells were incubated for 4 h. Formazan (100 μL) was added and continued to incubate until dissolved. The OD value at 570 nm was measured by a microplate reader (800TS, BIOTEK, USA).

Cell Viability Assay for IC50 Determination

Check if the same lab product or an alternative is used in the 5 most similar protocols
MTT detection kit (Beyotime, Shanghai, China) was employed to detect cell viability and determine the half maximal inhibitory concentration (IC50) of SEV. In short, NCM cells, SW620 cells or SE480 cells were grown in 96-well plate for 18 h and treated with the desired treatments. After 24 h, cells were incubated with MTT solution (Beyotime) for 4 h in an incubator. Then, dimethyl sulfoxide (Millipore) was incubated with cells at 37°C for 4 h. Cell viability was determined by analyzing the absorbance at 570 nm using a microplate reader (Thermo Labsystems, Waltham, MA, USA). The concentration of SEV causing 50% inhibition of growth (IC50) was determined by assessing the cell viability.

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
