In Vivo Tumor Growth and Metastasis

Female C57BL/6J mice aged 6–8 weeks old (15–20 g) were purchased from the Vital River Laboratories (Beijing, China). Protocol and procedures employed were reviewed and approved for handling mice and were approved by the Ethics Committee of Xinxiang Medical University. For examination of tumor growth in vivo, 12 mice were randomly divided into two groups and subcutaneously injected with 1 × 106 B16F10 cells bearing control or PRDM5 siRNA plasmids. Mice were killed 11 days later and the tumor size was determined. For examination of tumor metastasis, single cell suspension (1 × 106) in 100 μL of PBS was injected via the tail vein, and the mice were killed 2 weeks later. All the organs were examined for the macroscopic metastases. Lung metastatic nodules were determined under a dissecting microscope.

Modeling Colorectal Cancer in Mice

Female C57BL/6J mice aged 6–8 weeks were purchased from Beijing Vital River Laboratory Animal Technology Co., Ltd (Beijing, China). All animals were housed under controlled SPF conditions (temperature 22 ± 1°C, 12 h dark/light cycle) with free access to a standard diet and water. The Animal Ethics Committee approved animal experiments of the First Affiliated Hospital of Harbin Medical University. All experimental procedures were performed in accordance with the Guidelines for Ethical review of laboratory animal welfare (GB/T 35892–2018). A single intraperitoneal injection of AOM (Sigma-Aldrich, St. Louis, MO, USA) (12 mg/kg) was given to mice, and DSS (MP Biomedicals, California, USA) (2%) was given three rounds, one week each round, and free water was given two weeks after each round of DSS. Mice in the experimental group were given F. nucleatum by gavage (1.0×109 CFU/100μL) from two weeks before modeling until sacrifice. The control group was given the same amount of PBS by gavage, and the gavage was stopped during DSS. The mice were given broad-spectrum antibiotics (BSA) one week before gavage, which ensured the consistency of the intestinal flora of all mice. The body weight and disease activity index (DAI) changes of mice were monitored daily.

The regimen is designed based on our previous findings, which show that increasing DNA dosage for the 3rd vaccination can ensure the efficiency of in vivo antigen expression (30 (link)). The control group was boosted with PBS. Four weeks after the final vaccination, the mice were euthanized. Peripheral blood and splenocytes were collected for assays of S protein specific immune responses.
