DHW-221 was synthesized and purified as described previously (23 (link)). GDC-0980 and MG132 were obtained from Bide Pharmtech Ltd. (Shanghai, China). Taxol, docetaxel, vincristine, cisplatin, verapamil, and cycloheximide (CHX) were purchased from MedChem Express (NJ, USA). Rhodamine (Rho-123) and crystal violet were obtained from Shanghai Maclin Biochemical Co., Ltd. (Shanghai, China). Hoechst 33342 and 4,6-diamino-2-phenyl indole (DAPI) were obtained from Beyotime Biotechnology (Nanjing, China). The Annexin-V fluorescein isothiocyanate (FITC)/PI-double staining apoptosis detection kit and propidium iodide (PI) were obtained from Becton, Dickinson and Company (NJ, USA). P-gp antibody (CST, #13978; Rabbit, AF5185), PI3Kp110α antibody (rabbit, AF5112), p-FOXO3a (Ser253) antibody (rabbit, AF3020), and Bim antibody (Rabbit, AF0121) were purchased from Affinity Biosciences Pty Ltd. (Melbourne). Lactic dehydrogenase (LDH) detection kit (WL03271) and other primary antibodies were purchased from Wanlei Biotechnology Co., Ltd. (Shenyang, China)

Chemotherapeutic Drugs Evaluation Protocol

Chemotherapeutic drugs, VP-16, DXR, PTX, CDDP, carboplatin, gemcitabine, 5-FU, cyclophosphamide, capecitabine, and vincristine were purchased from MedChemExpress. Topo IIα (catalog 12286), Smurf2 (catalog 12024), cPARP (catalog 5625), and Bim (catalog 2933) antibodies were purchased from Cell Signaling Technology Inc. Mcl-1 (sc-12756) and Topo IIβ (sc-55330) antibodies were purchased from Santa Cruz Biotechnology. Anti–phospho-histone H2AX (Ser139; γ-H2AX) antibody was purchased from MilliporeSigma (catalog 05–636). DAPI (catalog 62248) and secondary antibody Alexa Fluor 488 donkey anti-mouse (A32766) were purchased from Thermo Fisher Scientific. Other reagents and antibodies were the same as described previously (10 (link), 16 (link)).

Cytotoxicity Assay for Sarcoma Cell Lines

Between 1500 and 2000 cells per well of Plat-E, U2OS, SJSA1, HOS, and MG63 were seeded in a 384-well plate using a Multidrop Combi Reagent Dispenser (ThermoFisher Scientific) in 25 µL of media. Six thousand OHSU-SARC001 cells per well were seeded in a 96-well plate manually. Plat-E, U2OS, SJSA1, HOS, and MG63 were seeded using D10 media, whereas OHSU-SARC001 was seeded using DMEM:F12. Cells were cultured for 3–5 d depending on doubling time. Inhibitors (LY3023414, everolimus, bimiralisib, rapamycin, trametinib, doxorubicin [Selleckchem], vincristine [MedChemExpress)], actinomycin [MedChemExpress], DMSO, and staurosporine [Selleckchem]) were added using a D300 Digital Dispenser (Hewlett-Packard) ranging from 0.001 to 10 µM. Dose–response assays involving ipatasertib and afuresertib (MedChemExpress) with ranges of 0.001 to 10 µM were added manually to 96-well plates. Plat-E-, U2OS-, SJSA-, HOS-, and MG63-containing plates were incubated for 72 h at 37°C and 5% CO2. OHSU-SARC001 cells were incubated for 6 d (given slow doubling time) at 37°C, 5% CO2. Viability was measured using a Cell Counting Kit-8 (Bimake) and read on a Biotek Synergy 2 plate reader.

Screening Spectrum Collection Compounds for OATP1B1 Inhibition

Compounds from the Spectrum Collection, distributed in 80 wells of 96 well plates (100 nmol per well in 10 μl of DMSO), were screened for their inhibitory effectiveness against transport of E3S into CHO cells expressing OATP1B1; a total of 560 compounds were used. Each compound was diluted to a concentration of 20 μM, pH 7.4, to a final concentration of 2% dimethylsulfoxide (DMSO) using a VIAFLO multichannel electronic pipet (Integra Biosciences, Hudson, NH) 65 . The following compounds for prospective testing were purchased from MedChemExpress (Monmouth Junction, NJ): abamectin, asiaticoside, baloxavir, berbamine, bremelanotide, bromocriptine, cabazitaxel, doramectin, etoposide, lapatinib, mobocertinib, novobiocin, posaconazole, rifaximin, teniposide, umbralisib, vancomycin, vinblastine and vincristine. Pyronaridine tetraphosphate was purchased from BOC Sciences (Shirley, NY) and tilorone hydrochloride from Caymen Chemical Company (Ann Arbor, MI). These compounds were solubilized in DMSO at 20 mM prior to tested.
